目的 观察不同浓度的组蛋白去乙酰化酶抑制剂曲古抑菌素A(TSA)对口腔鳞癌细胞系Tca-8113细胞体外抑制作用及端粒酶的影响,从而可能提供TSA对口腔鳞癌临床化疗应用的实验依据.方法 采用不同浓度TSA对Tca-8113细胞进行药物干预,倒置显微镜观察细胞用药前后的形态学改变;MTT法检测细胞增殖活性,确定后续实验中TSA作用浓度及时间点;应用以PCR为基础的端粒重复序列扩增(TRAP-PCR)银染法检测端粒酶活性,用蛋白印迹(Western blot)方法检测端粒酶逆转催化亚基单位(hTERT)蛋白表达.结果 TSA呈时间剂量性抑制Tca-8113细胞的生长,同时它也能降低端粒酶活性及hTERT蛋白表达的变化趋势.结论 TSA能够抑制Tca-8113细胞的增殖,进而下调细胞端粒酶活性及hTERT蛋白表达,这可能是TSA抗肿瘤作用机制之一.

Objective To investigate the inhibitive effects of trichostation A (TSA) with different concentrations on oral cancer Tca-8113 cells and telomerase activity, and to provide experimental proof for the clinical applications of TSA in anticancer chemotherapy.Methods Tca-8113 cells were treated by TSA with different concentrations, and morphology of cells was examined by transmission electron microscope; proliferative activity of cells was assessed by using MTT so that the concentration of TSA and the time-point could be determined for the follow-up experiment; Telomerase activity was examined by using TRAP-PCR, and hTERT protein was analyzed by Western blotting.Results A time- and dose-dependent inhibition was detected in Tca-8113 treated by TSA. Both the telomerase activity and the expression level of hTERT decreased after TSA treatment.Conclusion The results indicated that TSA can effectively inhibit the proliferation of Tca-8113 cells and thus reduce the telomerase activity and expression level of hTERT, which might be one of the mechanisms for the anticancer function of TSA.
