Small ubiquitin-related modifier (SUMO)4 is a newly found member of SUMO family. The biochemical effects of SUMO are somehow similar with those of ubiquitin, but SUMO stabilizes proteins, not degrades. The effect of SUMO4 is possibly mediated by NF-kB,with its antagonistic effect to ubiquitin. SUMO4 M55V mutation could repress sumoylation, activate ubiquitin and NF-kB, which leads to immunolog-ical responses and inflammation. It has already been verified that the polymorphism of M55V in SUMO4 is as-soeiated with type 1 diabetes in some populations. Some ongoing researches will try to explore its role in the pathogenesis of type 2 diabetes.
