[目的]建立天胡荽及其近缘植物的槲皮素HPLC含量分析方法,为进一步研究天胡荽及其近缘植物的药效学奠定基础.[方法]以槲皮素为指标成分,采用高效液相色谱法,以Hypersil ODS(4.6 mm×200 mm,5 μm)为色谱柱,甲醇-0.4%磷酸溶液(60∶40)为流动相,流速为1.0 ml/min,检测波长为368 nm,柱温为30 ℃,外标法进行测定.[结果]槲皮素进样量在0.081 6~0.571 2 μg范围内呈良好的线性关系,回归方程为y=145 361x+42 948,R~2=0.999 4(n=7);槲皮素加样回收率为100.48%,RSD为2.03%,含量为:天胡荽(0.081 6%)>破铜钱(0.078 1%)>柄花天胡荽(0.073 2%)>尼泊尔天胡荽(0.063 2%)>肾叶天胡荽(0.045 3%).[结论]建立的用HPLC法测定天胡荽及其近缘种中槲皮素含量的方法稳定、重复性好,为天胡荽及其近缘植物的化学组分与药效学的相关性研究提供了参考依据.
[目的]建立天鬍荽及其近緣植物的槲皮素HPLC含量分析方法,為進一步研究天鬍荽及其近緣植物的藥效學奠定基礎.[方法]以槲皮素為指標成分,採用高效液相色譜法,以Hypersil ODS(4.6 mm×200 mm,5 μm)為色譜柱,甲醇-0.4%燐痠溶液(60∶40)為流動相,流速為1.0 ml/min,檢測波長為368 nm,柱溫為30 ℃,外標法進行測定.[結果]槲皮素進樣量在0.081 6~0.571 2 μg範圍內呈良好的線性關繫,迴歸方程為y=145 361x+42 948,R~2=0.999 4(n=7);槲皮素加樣迴收率為100.48%,RSD為2.03%,含量為:天鬍荽(0.081 6%)>破銅錢(0.078 1%)>柄花天鬍荽(0.073 2%)>尼泊爾天鬍荽(0.063 2%)>腎葉天鬍荽(0.045 3%).[結論]建立的用HPLC法測定天鬍荽及其近緣種中槲皮素含量的方法穩定、重複性好,為天鬍荽及其近緣植物的化學組分與藥效學的相關性研究提供瞭參攷依據.

[Objective] The research aimed to establish a stable method for the content determination of quercetin in Hydrocotyle sibthorpioides Lam. And closely related species and lay a foundation for the further study of their pharmacodynamics.[Method] Using quercetin as the index compisition, HPLC was performed using Hypersil ODS C18 column (4.6 mm×200 mm,5 μm) and methanol-0.4% phosphoric acid(60∶40) as the mobile phase. The flow rate was 1.0 ml/min, the detection wavelength was 368 nm, and the column temperature was 30 ℃.The external standard method was used as quantitative analysis. [Result] The linear range of Quercetin was 0.081 6-0.571 2 g, the regression equation was Y=145 361X+42 948, r=0.999 4(n=7); the average recovery was 100.48%, RSD=2.03%. The results showed as follows: H. Sibthorpioides of 0.081 6%, H. Sibthorpioides var. Batrachium of 0.078 1%, H. Podantha of 0.0732%, H. Nepalensis of 0.063 2% and H. Wilfordi of 0.045 3%. [Conclusion] The method is stable and with good repeatability. This research provides a helpful reference for the research of the correlation between chemical components and the pharmacodynamics in Hydrocotyle sibthorpioides and closely related species.
