目的:观察骨形态发生蛋白(bone morphogenetic proteins,BMPs)及其受体在正常成年大鼠脊髓中的表达分布.方法::取正常成年大鼠脊髓并切片,运用免疫组织化学方法分别检测BMP2,4,7及其受体BMPR Ⅰa,BMPR Ⅰb,BMP Ⅱ的表达.结果:在正常成年大鼠脊髓的前角运动神经元、背角感觉神经元、少突胶质细胞、部分小胶质细胞和部分星形胶质细胞表达BMPR Ⅰa和BMPR Ⅰb.少突胶质细胞及灰质前角运动神经元表达BMPR Ⅱ受体,灰质神经胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)阳性的星形胶质细胞未见BMPR Ⅱ受体表达,白质的部分GFAP阳性的星形胶质细胞表达BMPR Ⅱ受体.免疫组织化学未检测出BMP2,4在正常成年大鼠脊髓中表达.BMP7在所有的APC阳性的少突胶质细胞及NeuN阳性的前角运动神经元中表达,在正常脊髓的部分星形胶质细胞中可见表达.磷酸化的pSmad 1/5/8蛋白表达见于所有的少突胶质细胞及神经元、部分星形胶质细胞以及软膜下脊髓周边GFAP阳性的星形胶质细胞,这些细胞也是RC2阳性细胞(即放射状胶质细胞).结论:BMP2,4在正常成年大鼠脊髓中不表达,BMP受体、BMP7及磷酸化的pSmad 1/5/8蛋白在正常成年大鼠脊髓的多种细胞中表达,提示BMP信号在成体脊髓神经元及胶质细胞功能中起重要作用.
目的:觀察骨形態髮生蛋白(bone morphogenetic proteins,BMPs)及其受體在正常成年大鼠脊髓中的錶達分佈.方法::取正常成年大鼠脊髓併切片,運用免疫組織化學方法分彆檢測BMP2,4,7及其受體BMPR Ⅰa,BMPR Ⅰb,BMP Ⅱ的錶達.結果:在正常成年大鼠脊髓的前角運動神經元、揹角感覺神經元、少突膠質細胞、部分小膠質細胞和部分星形膠質細胞錶達BMPR Ⅰa和BMPR Ⅰb.少突膠質細胞及灰質前角運動神經元錶達BMPR Ⅱ受體,灰質神經膠質纖維痠性蛋白(glial fibrillary acidic protein,GFAP)暘性的星形膠質細胞未見BMPR Ⅱ受體錶達,白質的部分GFAP暘性的星形膠質細胞錶達BMPR Ⅱ受體.免疫組織化學未檢測齣BMP2,4在正常成年大鼠脊髓中錶達.BMP7在所有的APC暘性的少突膠質細胞及NeuN暘性的前角運動神經元中錶達,在正常脊髓的部分星形膠質細胞中可見錶達.燐痠化的pSmad 1/5/8蛋白錶達見于所有的少突膠質細胞及神經元、部分星形膠質細胞以及軟膜下脊髓週邊GFAP暘性的星形膠質細胞,這些細胞也是RC2暘性細胞(即放射狀膠質細胞).結論:BMP2,4在正常成年大鼠脊髓中不錶達,BMP受體、BMP7及燐痠化的pSmad 1/5/8蛋白在正常成年大鼠脊髓的多種細胞中錶達,提示BMP信號在成體脊髓神經元及膠質細胞功能中起重要作用.
목적:관찰골형태발생단백(bone morphogenetic proteins,BMPs)급기수체재정상성년대서척수중적표체분포.방법::취정상성년대서척수병절편,운용면역조직화학방법분별검측BMP2,4,7급기수체BMPR Ⅰa,BMPR Ⅰb,BMP Ⅱ적표체.결과:재정상성년대서척수적전각운동신경원、배각감각신경원、소돌효질세포、부분소효질세포화부분성형효질세포표체BMPR Ⅰa화BMPR Ⅰb.소돌효질세포급회질전각운동신경원표체BMPR Ⅱ수체,회질신경효질섬유산성단백(glial fibrillary acidic protein,GFAP)양성적성형효질세포미견BMPR Ⅱ수체표체,백질적부분GFAP양성적성형효질세포표체BMPR Ⅱ수체.면역조직화학미검측출BMP2,4재정상성년대서척수중표체.BMP7재소유적APC양성적소돌효질세포급NeuN양성적전각운동신경원중표체,재정상척수적부분성형효질세포중가견표체.린산화적pSmad 1/5/8단백표체견우소유적소돌효질세포급신경원、부분성형효질세포이급연막하척수주변GFAP양성적성형효질세포,저사세포야시RC2양성세포(즉방사상효질세포).결론:BMP2,4재정상성년대서척수중불표체,BMP수체、BMP7급린산화적pSmad 1/5/8단백재정상성년대서척수적다충세포중표체,제시BMP신호재성체척수신경원급효질세포공능중기중요작용.
Objective To observe the expression distribution of bone morphogenetic proteins (BMP) in the spinal cord of normal adult rats. Methods Expression of BMP2, BMP4, and BMP7, and their receptors BMPR Ⅰa, BMPR Ⅰb, and BMP Ⅱ were detected by immunochemistry analysis in the spinal cord of normal adult rats. Results Expression of BMPR Ia or BMPR Ib was observed in the motor neurons of the anterior horn, sensory neurons of the dorsal horn, oligodentrocytes, some microglia, and some astrocytes. Expression of receptor BMPR Ⅱ was found in the oligodentrocytes and motor neurons in the gray matter of anterior horn. It was also expressed in some glial fibrillary acidic protein (GFAP)-positive astrocytes in the white matter but not in the gray matter. BMP2 and BMP4 were not expressed in the spinal cord of normal adult rats by immunohistochemistry. BMP7 was expressed in all the APC-positive oligodentrocytes, all the NeuN-positive motor neurons in the anterior horn, and some astrocytes in the normal spinal cord. Phosphated pSmad 1/5/8 protein was expressed in all the oligodentrocytes, all the neurons, and some astrocytes, especially in the GFAP-positive astrocytes which were RC2-positive radial glia in the subventricular zone.Conclusion BMP7, BMP receptors, and phosphated pSmad 1/5/8 are expressed in many types of cells whereas BMP2 and BMP4 are not expressed in the spinal cord of normal adult rats, which suggests an important function of BMP signal pathway in the neuron and glia of spinal cord.
