目的:探讨脑缺血早期(24h内)大脑皮质星形胶质细胞(Ast)的变化规律。方法:应用胶质原纤维性酸性蛋白免疫组织化学ABC技术。结果:缺血15 min和30min组,缺血中心区大脑皮质胶质原纤维性酸性蛋白阳性细胞较均匀地分布于Ⅱ~Ⅵ层,细胞数量明显多于非缺血区;缺血1 h和2 h组胶质原纤维性酸性蛋白阳性细胞数量进一步增加,胞质染色加深,并集中出现于Ⅲ、Ⅵ层;缺血3 h组胶质原纤维性酸性蛋白阳性细胞进一步增大呈气球样,突起增长变粗,突起内出现水肿泡;缺血6 h组胶质原纤维性酸性蛋白细胞周缩,边界不清;12 h和24h组缺血中心区胶质原纤维性酸性蛋白细胞消失。同时观察到缺血3 h和6 h组缺血边缘区胶质原纤维性酸性蛋白细胞数量增多,直径增大,并可见到细胞分裂现象。结论:星形胶质细胞对脑缺血早期神经元损伤具有较强的保护作用。

목적:탐토뇌결혈조기(24h내)대뇌피질성형효질세포(Ast)적변화규률。방법:응용효질원섬유성산성단백면역조직화학ABC기술。결과:결혈15 min화30min조,결혈중심구대뇌피질효질원섬유성산성단백양성세포교균균지분포우Ⅱ~Ⅵ층,세포수량명현다우비결혈구;결혈1 h화2 h조효질원섬유성산성단백양성세포수량진일보증가,포질염색가심,병집중출현우Ⅲ、Ⅵ층;결혈3 h조효질원섬유성산성단백양성세포진일보증대정기구양,돌기증장변조,돌기내출현수종포;결혈6 h조효질원섬유성산성단백세포주축,변계불청;12 h화24h조결혈중심구효질원섬유성산성단백세포소실。동시관찰도결혈3 h화6 h조결혈변연구효질원섬유성산성단백세포수량증다,직경증대,병가견도세포분렬현상。결론:성형효질세포대뇌결혈조기신경원손상구유교강적보호작용。
Objective: To clarifiy the changes of the astrocytes (Ast) during early stage (within 24 hours) of focal cerebral ischemia. Methods: Using ABC immunohistochemical technique for GFAP. Results:GFAP-positive cells were evenly distributed between layers of Ⅱ~ Ⅵ of ischemia foci in the cerebral cortex in 15 min and 30 rnin ischemia groups, and the numbers of which were more than those in normal cerebral regions. In 1 hour and 2 hours groups, the numbers of GFAP-positive cells increased,staining of their cytoplasms were darker, ahd concentrated at layers Ⅲ and Ⅳ. In 3 hours group, the GFAP-positive cells became a balloon-like in shape, and their processes enlarged and lengthened farther, and some edema blebs emerged. As for 6 hour group, the GFAP-positive cells were shrunken with obscure contours. In 12 hours and 24 hours groups, the GFAP-positive cells disappeared. At ischemia border zone, the GFAP-positive cells increased in number and cell division appeared in 3 hours and 6 hours groups. Conclusion: Astrocytes play a protective role for damaged neurons at early stage during cerebral ischemia.
