利用热激PCR法和菌落直接PCR法进行大肠杆菌和农杆菌的鉴定,筛选含有GFP-linker-C-TAPa或wtGPA1-C-TAPa的pCAMBIA2300EC的阳性克隆,比较热激PCR法和菌落直接PCR法的优劣.热激PCR法阳性检出率是100%,菌落直接PCR法阳性检出率是60%左右.热激PCR法是一种快速可靠的鉴定重组阳性克隆的方法.
利用熱激PCR法和菌落直接PCR法進行大腸桿菌和農桿菌的鑒定,篩選含有GFP-linker-C-TAPa或wtGPA1-C-TAPa的pCAMBIA2300EC的暘性剋隆,比較熱激PCR法和菌落直接PCR法的優劣.熱激PCR法暘性檢齣率是100%,菌落直接PCR法暘性檢齣率是60%左右.熱激PCR法是一種快速可靠的鑒定重組暘性剋隆的方法.

To compare advantage and disadvantage of microbe screen between heat shock PCR method and colony direct PCR method,the positive clones of pCAMBIA2300EC with GFP-linker-C-TAPa or wtGPAI-C-TAPa were screened by heat shock PCR method and colony direct PCR method in Escherichia coil and Agrobacterium tumefaciens respectively.The positive rates of screening clones by heat shock PCR method were 100%.The positive rates of screening clones by colony direct PCR method were about 60%.Result showed that heat shock PCR method is effective way for microbe screen.
