目的 观察粒细胞集落刺激因子(G-CSF)对心肌梗死后动作电位的影响.方法 38只大白兔开胸结扎左冠脉前降支,治疗组皮下注射G-CSF每天10μ/kg连续5d,对照组皮下注射每天0.5 ml/kg生理盐水连续5d;观察心肌细胞不同部位不同时间心肌动作电位的变化.结果 在术后30 min、7d、14d、30d、60d两组心肌动作电位在中央区、移行区及边缘区均有下降,但治疗组在移行区较快恢复[治疗组7d (120.0±3.9) ms、14d (136.0±3.3) ms、30d (140.0±4.7) ms;对照组7d (107.0±4.4) ms、14d (112.0±4.6) ms、30d (120.0±7.1) ms,P<0.05];左室射血分数(LVEF)治疗组(70.3±9.4)%较对照组(58.2±13.7)%明显升高(P<0.05),短轴缩短率(FS)治疗组(39.2±6.4)%较对照组(28.711.1)%明显升高(P<0.05).结论 G-CSF能够加速心肌梗死后心肌动作电位的恢复;从而有效地加速疾病恢复的进程.

Objective To observe the effects of granulocyte colony-stimulating factors (G-CSF)on action potential of cardiac myocytes in a rabbit model of acute myocardial infarction.Methods Under anesthesia and mechanical ventilation with room air,a left thoracotomy was performed,and 4-0 silk string was used to ligate the left branch of coronary artery.Thirty-eight healthy New Zealand white rabbits were divided into two groups:G-CSF group and normal saline group.10 μg/(kg·day) G-CSF in G-CSF group and 0.5 ml/(kg·day) normal saline in control group were given 24 h after infarction,one time per day for 5 days.The changes in action potential of cardiac myocytes at different areas and time points after acute myocardial infarction were observed.Results Action potential duration of cardiac myocytes was obviously decreased 30 min,7 days,14 days,30 days,and 60 days after myocardial infarction in different areas in both two groups,but recovered fastly in treatment group [7th day:( 120.0 ±3.9) ms,14th day:( 136.0 ±3.3) ms,30th day:(140.0 ±4.7) ms]as compared with control group in marginal area [7th day:(107.0 ±4.4) ms,14th day:(112.0 ±4.6) ms,30th day:(120.0 ±7.1) ms],P<0.05.Ejection fraction and fractional shortening were significantly increased in treatment group as compared with control group ( both P < 0.05 ).Conclusion The change of action potential suggests G-CSF may play an important role in the treatment of myocardial infarction.
