目的 观察血小板衍生生长因子(PDGF)A、B及其α,β受体在大鼠野百合碱肺高压模型中的表达,探讨PDGF在肺血管重构中的作用.方法 SD大鼠24只,体质量250~300g,随机分为野百合碱注射组(MCT,12只)和对照组(Con,12只).MCT组给与野百合碱60 mg/kg单次皮下注射.喂养6周后,使用逆转录-聚合酶链反应(RT-PCR)观察肺组织PDGF-A、PDGF-B及其α、β受体mRNA转录量的变化.结果 MCT组肺组织中PDGF-A、PDGF-B、PDGFR-α、PDGFR-β的mRNA转录水平都高于对照组(1.5609±0.1246比0.4057±0.0007,P<0.05;0.4810±0.0733比0.2639±0.0070,P<0.05:0.4963±0.0137比0.3038±0.0859,P<0.05;1.1257±0.2490比0.0736±0.0185,P<0.05).结论 血小板衍生生长因子(PDGF)-A、PDGF-B及其α、β受体活性表达的增强,参与了肺动脉高压肺血管重构的机制.
目的 觀察血小闆衍生生長因子(PDGF)A、B及其α,β受體在大鼠野百閤堿肺高壓模型中的錶達,探討PDGF在肺血管重構中的作用.方法 SD大鼠24隻,體質量250~300g,隨機分為野百閤堿註射組(MCT,12隻)和對照組(Con,12隻).MCT組給與野百閤堿60 mg/kg單次皮下註射.餵養6週後,使用逆轉錄-聚閤酶鏈反應(RT-PCR)觀察肺組織PDGF-A、PDGF-B及其α、β受體mRNA轉錄量的變化.結果 MCT組肺組織中PDGF-A、PDGF-B、PDGFR-α、PDGFR-β的mRNA轉錄水平都高于對照組(1.5609±0.1246比0.4057±0.0007,P<0.05;0.4810±0.0733比0.2639±0.0070,P<0.05:0.4963±0.0137比0.3038±0.0859,P<0.05;1.1257±0.2490比0.0736±0.0185,P<0.05).結論 血小闆衍生生長因子(PDGF)-A、PDGF-B及其α、β受體活性錶達的增彊,參與瞭肺動脈高壓肺血管重構的機製.
목적 관찰혈소판연생생장인자(PDGF)A、B급기α,β수체재대서야백합감폐고압모형중적표체,탐토PDGF재폐혈관중구중적작용.방법 SD대서24지,체질량250~300g,수궤분위야백합감주사조(MCT,12지)화대조조(Con,12지).MCT조급여야백합감60 mg/kg단차피하주사.위양6주후,사용역전록-취합매련반응(RT-PCR)관찰폐조직PDGF-A、PDGF-B급기α、β수체mRNA전록량적변화.결과 MCT조폐조직중PDGF-A、PDGF-B、PDGFR-α、PDGFR-β적mRNA전록수평도고우대조조(1.5609±0.1246비0.4057±0.0007,P<0.05;0.4810±0.0733비0.2639±0.0070,P<0.05:0.4963±0.0137비0.3038±0.0859,P<0.05;1.1257±0.2490비0.0736±0.0185,P<0.05).결론 혈소판연생생장인자(PDGF)-A、PDGF-B급기α、β수체활성표체적증강,삼여료폐동맥고압폐혈관중구적궤제.
Objective To investigate the expression of PDGF-A,PDGF-B,PDGFR-α,PDGFR-β mRNA in lung of rat model of pulmonary hypertension induced by injection of monocrotaline and their role in pulmonary vascular remodeling.Methods Twenty-four SD rats,weighing 250-300 g,were randomly divided into 2 groups:monocrotaline group(MCT,n=12),control group(Con,n=12).111e rats in MCT group received a single subcutaneous injection of monocrotaline(60 ms/kg).After fed for 6 weeks,the expression of PDGF-A,PDGF-B,PDGFR-α,and PDGFR-β mRNA in lung tissue was detected by RT-PCR.Results In MCT group,the expression levels of PDGF-A,PDGF-B,PDGFR-ctand PDGFR-β mRNA were higher than those in control group(1.5609±0.1246 vs 0.4057±0.0007,P<0.05:0.4810±0.0733 vs 0.2639±0.0070.P<0.05;0.4963±0.0137 vs 0.3038±0.0859,P<0.05;1.1257±0.2490 vs 0.0736±0.0185.P<0.05).Conclusion The higher expression level of PDGF-A.PDGF-B,PDGF R-α,and PDGFR-β may involve in the mechanism of pulmonary vascular remodeling in rat model of pulmonary hypertension induced by injection of monocrotaline.
