目的 观察家蝇幼虫和蛹抗菌肽对弓形虫速殖子的抑制作用. 方法 采用损伤加感染的方法,诱导家蝇幼虫和蛹大量产生抗菌肽,通过研磨、低温高速离心等方法获得抗菌肽粗提物,分别用磷酸缓冲液(PBS)调整为4 mg/ml、2 mg/ml和1 mg/ml并作用于弓形虫速殖子,24 h后,通过血细胞计数法和扫描电镜观察两种粗提物对弓形虫速殖子的抑制作用. 结果 幼虫抗菌肽组和蛹抗菌肽组速殖子存活率均低于对照组,差异有统计学意义(P<0.05),其抗弓形虫作用随着抗菌肽浓度的减小而减小;扫描电镜观察,对照组弓形虫速殖子外型完整,表面平滑,抗菌肽组速殖子则虫体膨胀.表面凹凸不平. 结论 从家蝇幼虫和蛹中提取的抗菌肽都有抗弓形虫作用,幼虫抗菌肽效果更好.

목적 관찰가승유충화용항균태대궁형충속식자적억제작용. 방법 채용손상가감염적방법,유도가승유충화용대양산생항균태,통과연마、저온고속리심등방법획득항균태조제물,분별용린산완충액(PBS)조정위4 mg/ml、2 mg/ml화1 mg/ml병작용우궁형충속식자,24 h후,통과혈세포계수법화소묘전경관찰량충조제물대궁형충속식자적억제작용. 결과 유충항균태조화용항균태조속식자존활솔균저우대조조,차이유통계학의의(P<0.05),기항궁형충작용수착항균태농도적감소이감소;소묘전경관찰,대조조궁형충속식자외형완정,표면평활,항균태조속식자칙충체팽창.표면요철불평. 결론 종가승유충화용중제취적항균태도유항궁형충작용,유충항균태효과경호.
Objective To investigate the anti-Toxoplasma activity of the antimicrobial peptides(AMPs) extracted from the larvae and pupae of the housefly. Methods The crude AMPs were extracted by trituration and centrifugation from the larvae and pupae induced by intection and injury,then adjusted to 4 mg/ml,2 mg/ml and 1 mg/ml by phosphate buffered solution (PBS) respectively, then added into the suspension of tachyzoites of Toxoplasma gondii. The anti-Toxoplasma effect of AMPs were observed by the cell counting method and scanning electron microscope (SEM) at 24 h of incubation. Results It was found that the survival rate of tachyzoites in the larva group and pupae group was lower than that of the control group (P<0.05). And the anti-Toxoplasma effect decreased along with the reduction of protein concentration in both groups. From SEM, it was found that the cellular membrane of the tachyzoites in the larva group was rugged and swollen compared with that in the control group, of which the cellular membrane was integrated and smooth. Conclusion AMPs extracted from the larvae and pupae of housefly had anti-Toxoplasma aetivity,and the larva AMPs had a better effect.
