目的 探讨游离脂肪酸功能受体G蛋白偶联受体120(GPR120)在肥胖患者腹部皮下脂肪组织和大网膜脂肪组织中mRNA表达情况及其相关的影响因素.方法 于2010年9月至2011年8月选取广州医学院第三附属医院施行慢性胆囊炎择期手术的非超重患者[对照组,体质指数(BMI)<23 kg/m2] 39例和肥胖患者(肥胖组,BMI≥25 kg/m2)40例,测量体重、腰臀围、血糖、血脂等指标,用逆转录-聚合酶链反应法(RT-PCR)检测腹部大网膜与皮下脂肪组织GPR120 mRNA的表达水平.采用t检验、Pearson相关分析、偏相关分析和多元逐步回归进行统计学分析.结果 (1)大网膜组织中的GPR120 mRNA表达在肥胖组(0.876±0.076)和对照组(0.621±0.052)均高于两组皮下脂肪组织中的水平(0.479±0.028、0.421±0.035),差异有统计学意义(t值分别为-2.546、-2.211,均P<0.05).肥胖组大网膜的GPR120 mRNA表达量(0.876±0.076)高于对照组(0.621±0.052),差异有统计学意义(t=-2.348,P<0.05),皮下脂肪GPR120 mRNA表达量在肥胖组(0.479±0.028)与对照组(0.421±0.035)之间差异无统计学意义(t=-0.584,P>0.05).(2)以大网膜脂肪组织GPR120 mRNA表达量为应变量,仅稳态模型胰岛素抵抗指数和腰臀比进入分析模型,所得模型R2=0.3730,标准偏回归系数分别为0.227和0.321.皮下脂肪GPR120 mRNA表达量与年龄、血酯等其他因素均无相关性(均P >0.05).结论 肥胖患者腹部大网膜脂肪组织GPR120 mRNA表达明显升高,腹型肥胖、胰岛素抵抗是大网膜脂肪组织GPR120表达水平的主要影响因素.

목적 탐토유리지방산공능수체G단백우련수체120(GPR120)재비반환자복부피하지방조직화대망막지방조직중mRNA표체정황급기상관적영향인소.방법 우2010년9월지2011년8월선취엄주의학원제삼부속의원시행만성담낭염택기수술적비초중환자[대조조,체질지수(BMI)<23 kg/m2] 39례화비반환자(비반조,BMI≥25 kg/m2)40례,측량체중、요둔위、혈당、혈지등지표,용역전록-취합매련반응법(RT-PCR)검측복부대망막여피하지방조직GPR120 mRNA적표체수평.채용t검험、Pearson상관분석、편상관분석화다원축보회귀진행통계학분석.결과 (1)대망막조직중적GPR120 mRNA표체재비반조(0.876±0.076)화대조조(0.621±0.052)균고우량조피하지방조직중적수평(0.479±0.028、0.421±0.035),차이유통계학의의(t치분별위-2.546、-2.211,균P<0.05).비반조대망막적GPR120 mRNA표체량(0.876±0.076)고우대조조(0.621±0.052),차이유통계학의의(t=-2.348,P<0.05),피하지방GPR120 mRNA표체량재비반조(0.479±0.028)여대조조(0.421±0.035)지간차이무통계학의의(t=-0.584,P>0.05).(2)이대망막지방조직GPR120 mRNA표체량위응변량,부은태모형이도소저항지수화요둔비진입분석모형,소득모형R2=0.3730,표준편회귀계수분별위0.227화0.321.피하지방GPR120 mRNA표체량여년령、혈지등기타인소균무상관성(균P >0.05).결론 비반환자복부대망막지방조직GPR120 mRNA표체명현승고,복형비반、이도소저항시대망막지방조직GPR120표체수평적주요영향인소.
Objective To examine the expression of G-protein coupled receptor 120 (GPR120) mRNA in abdominal subcutaneous and omental adipose tissues and related factors in obese subjects.Methods From September 2010 to August 2011,39 non overweight subjects ( body mass index ( BMI )<23 kg/m2) and 40 obese patients (BMI > 25 kg/m2) who underwent cholecystectomy in the Third Affiliated Hospital of Guangzhou Medical College were enrolled in this study. Body weight,waist-hip ratio (WHR),blood glucose,and lipids,etc,were measured.The expression of GPR120 mRNA in abdominal subcutaneous and omental adipose tissues were measured by RT-PCR. The t test,Pearson correlation analysis,partial correlation analysis and muhi-stepwise regression were used for data analysis. Results ( 1 ) In obese and control group,the GPR120 mRNA expression in omental adipose tissue (0.876 ±0.076,0.621 ± 0.052 ) were significantly higher than those in subcutaneous adipose tissue ( 0.479 ± 0.028,0.421 ± 0.035,t =-2.546 or -2.211,respectively,both P < 0.05 ).The GPR120 mRNA expression in omental adipose tissue (0.876 ± 0.076 ) was significantly higher in obese group than that in control group (0.621 ± 0.052,t =- 2.348,P < 0.05 ). The differences in the expression of GPR120 mRNA in abdominal subcutaneous adipose tissue between the two groups were not significant (0.479 ±0.028 vs 0.421 ±0.035,t =0.584,P > 0.05 ). (2) Multi-stepwise regression analysis demonstrated that the expression of GPR120 mRNA in omental adipose tissue was significantly correlated with HOMA-IR and WHR (R2 =0.3730,standard partial regression coefficient were 0.227 and 0.321,respectively).The expression of GPR120 mRNA in abdominal subcutaneous adipose tissue was not correlated with other factors.Conclusion The GPR120 mRNA expression in abdominal omental adipose tissue was significantly higher in obese subjects than that in control group. Abdominal obesity and HOMA-IR were the main independent factors influencing the expression of GPR120 mRNA in omental adipose tissue.
