采用弱酸处理法,从新鲜牛皮中提取了胶原蛋白,通过与明胶对比,采用CD、AFM、FTIR和荧光方法对其结构形态进行了表征研究.结果表明,所提取胶原蛋白保持了固有的生物活性,具有典型的α-螺旋结构.首次建立了稳态荧光的表征方法,不仅与其他方法具有重要互补性,而且具有准确、灵敏、快捷的特点,是表征、研究胶原蛋白及其变性/复性行为的有效手段.

The weak acid method was used to prepare collagen from fresh bovine skin. The conformational structure of the collagen was studied by CD, AFM, FTIR and fluorescence methods via comparison with that of gelatin. The results show that our product keeps the natural collagen typical structure of α-helix. Especially, we have established the characterization method of steady state fluorescence for the first time, which, to the existing technique, is not only an important complementary method with features of being accurate, sensitive and fast, but also a new powerful tool in studying the denaturation/renaturation behavior of collagen.
