为对死亡雏鸡进行病因诊断,通过大体剖检、细菌分离、生化鉴定,证实为铜绿假单胞菌感染。测定了该分离株的16SrRNA基因序列,并与GenBank中收录的序列比较,结果发现所分离的铜绿假单胞茵及参考株的16SrDNA基因序列极其保守,相似性达99%~100%;与大肠杆菌、沙门氏菌、巴氏杆菌的相似性差异约为9%;与鸭疫里默氏杆菌的相似性差异则更大,超过18%,提示16SrRNA序列测定可作为该菌鉴定的一种可靠方法。

Abstract. In this experiment a bacteria strain isolated fi'om one dead chicken was identified as Pseudomonas aeruginosa by morphological characteristics, cultural characteristics and biological characterization. The 16s rRNA gene sequence of this bacteria was also obtained by PCR with a pair of common primers. This strain was closely related to the strains of Pseudomonas aeruginosa after the 16s rRNA sequence of this strain was blasted in GenBank. The nucleotide sequence homology was from 99% to 100% between this strain and reference Pseudomonas aeruginosa strains. The discrepancy was about 9% between this strain and E.coli., Salmonella or Pasteurella strains. The discrepancy was more than 18% between this strain and Riemerella strains. These results showed that sequencing of 16S rRNA gene was a useful method for identifing the bacteria.
