目的:了解长春新碱(VCR)诱导的自噬性细胞凋亡是否与线粒体跨膜电位(ΔΨm )改变有关及其可能的机制。材料与方法:以正常人的肝细胞系为体外模型应用碘化丙啶(PI)/Rhodamine123(Rh123)双重染色检测ΔΨm,通过亚G1期细胞和透射电镜鉴定细胞凋亡。结果与结论:VCR可明显诱导线粒体ΔΨm下降,也可以诱导L-02细胞自噬性凋亡。线粒体ΔΨm下降可能是VCR诱导自噬性细胞凋亡的关键环节。
目的:瞭解長春新堿(VCR)誘導的自噬性細胞凋亡是否與線粒體跨膜電位(ΔΨm )改變有關及其可能的機製。材料與方法:以正常人的肝細胞繫為體外模型應用碘化丙啶(PI)/Rhodamine123(Rh123)雙重染色檢測ΔΨm,通過亞G1期細胞和透射電鏡鑒定細胞凋亡。結果與結論:VCR可明顯誘導線粒體ΔΨm下降,也可以誘導L-02細胞自噬性凋亡。線粒體ΔΨm下降可能是VCR誘導自噬性細胞凋亡的關鍵環節。
목적:료해장춘신감(VCR)유도적자서성세포조망시부여선립체과막전위(ΔΨm )개변유관급기가능적궤제。재료여방법:이정상인적간세포계위체외모형응용전화병정(PI)/Rhodamine123(Rh123)쌍중염색검측ΔΨm,통과아G1기세포화투사전경감정세포조망。결과여결론:VCR가명현유도선립체ΔΨm하강,야가이유도L-02세포자서성조망。선립체ΔΨm하강가능시VCR유도자서성세포조망적관건배절。
Purpose: To understand if vincristine (VCR) is related to the alteration of mitochondrial transmembrane potentials (ΔΨm ) and its possible mechanisms. Methods: Human liver cell line L-02 was used as in vitro models. The ΔΨm was detected by the double staining of propidium iodide (PI) and Rhodamine 123(Rh123),which fluorescent intensities were measured by flow cytometry, while apoptosis was confirmed by sub-G1 cell content as well as transmission electron microscopy observation. Results and Conclusion: In the cells sensitive to VCR-induced autophagic apoptosis, the ΔΨm was disrupted with VCR treatment. Vincristine can induced L-02 cell autophagic apoptosis, which could associate closely with the ΔΨm collapse. The disruption of the ΔΨm maybe is the key event of VCR-induced autophagic apoptosis.
