通过组织培养技术,利用胆木(Nauclea officinalis)种子无菌实生苗的茎段、叶片、叶柄等进行愈伤组织的诱导及悬浮细胞的培养。结果如下:(1)以WPM为基本培养基,获得愈伤组织高诱导率的激素种类及浓度为①0.5 mg/L 2,4-D、②0.5~1.5 mg/L IBA+(0.0,0.5 mg/L)6-BA,诱导率达到100%;(2)在外植体的差异上,茎段和叶片出现愈伤组织的时间较早、增殖系数最大,可达15倍;(3)在培养基1/2WPM+0.5 mg/L2,4-D+30 g/L蔗糖中,继代周期为14~17 d,胆木细胞悬浮培养增殖率可达30.16%。
通過組織培養技術,利用膽木(Nauclea officinalis)種子無菌實生苗的莖段、葉片、葉柄等進行愈傷組織的誘導及懸浮細胞的培養。結果如下:(1)以WPM為基本培養基,穫得愈傷組織高誘導率的激素種類及濃度為①0.5 mg/L 2,4-D、②0.5~1.5 mg/L IBA+(0.0,0.5 mg/L)6-BA,誘導率達到100%;(2)在外植體的差異上,莖段和葉片齣現愈傷組織的時間較早、增殖繫數最大,可達15倍;(3)在培養基1/2WPM+0.5 mg/L2,4-D+30 g/L蔗糖中,繼代週期為14~17 d,膽木細胞懸浮培養增殖率可達30.16%。
통과조직배양기술,이용담목(Nauclea officinalis)충자무균실생묘적경단、협편、협병등진행유상조직적유도급현부세포적배양。결과여하:(1)이WPM위기본배양기,획득유상조직고유도솔적격소충류급농도위①0.5 mg/L 2,4-D、②0.5~1.5 mg/L IBA+(0.0,0.5 mg/L)6-BA,유도솔체도100%;(2)재외식체적차이상,경단화협편출현유상조직적시간교조、증식계수최대,가체15배;(3)재배양기1/2WPM+0.5 mg/L2,4-D+30 g/L자당중,계대주기위14~17 d,담목세포현부배양증식솔가체30.16%。
Callus induction and suspension culture of Nauclea officinalis were conducted using stem segments,leaves,petioles,etc.of in vitro seedlings as explants.The results showed that the best combinations of plant growth regulators for callus induction on WPM were ①0.5 mg/L 2,4-D,and ②0.5-1.5 mg/L IBA+(0.0,0.5 mg/L) 6-BA,with frequencies of callus induction being 100%.The better explants were stem segments and leaves since calli appeared earlier on them and their multiplication coefficients were up to 15.The proliferation rate of cell suspension culture of Nauclea officinalis on 1/2WPM+0.5 mg/L 2,4-D+30 g/L sugar was up to 30.16 for a subculture period of about 14-17 days.
