目的 观察含母牛分枝杆菌菌苗(微卡)凝胶在肺部介入应用后产生的局部及全身免疫应答效应.方法 60只日本长耳兔随机分成5组,每组12只.第1组为对照组,气管插管至末梢支气管注入生理盐水3 ml;第2组为单纯凝胶组,肺部注入卡波姆凝胶3 ml;第3、4、5组为微卡凝胶组,分别注入含3.75、7.50、9.37 mg/L母牛分枝杆菌菌苗的卡波姆凝胶3 ml.持续观察4周,期间检测首次注入后与处死解剖前外周血T细胞亚群及γ-干扰素含量.结果 处死解剖前第4、5组外周血CD3+百分比[(55.8±9.7)%、(54.5±11.8)%]高于第1组[(43.1±3.0)%](P值均<0.05);第4、5组外周血CD4+百分比[(44.1±5.8)%、(43.3±6.2)%]高于第1组[(33.5±3.1)%](P值均<0.05);第3、4、5组外周血CD8+百分比[(21.3±3.3)%、(21.1±4.5)%、(20.5±4.1)%]低于第1组[(28.2±1.7)%](P值均<0.05).第4、5组检测微卡凝胶注入侧肺组织研磨混浆内γ-干扰素水平为阳性.结论 以健康日本长耳兔为模型局部免疫刺激可以引起全身免疫应答反应.

목적 관찰함모우분지간균균묘(미잡)응효재폐부개입응용후산생적국부급전신면역응답효응.방법 60지일본장이토수궤분성5조,매조12지.제1조위대조조,기관삽관지말소지기관주입생리염수3 ml;제2조위단순응효조,폐부주입잡파모응효3 ml;제3、4、5조위미잡응효조,분별주입함3.75、7.50、9.37 mg/L모우분지간균균묘적잡파모응효3 ml.지속관찰4주,기간검측수차주입후여처사해부전외주혈T세포아군급γ-간우소함량.결과 처사해부전제4、5조외주혈CD3+백분비[(55.8±9.7)%、(54.5±11.8)%]고우제1조[(43.1±3.0)%](P치균<0.05);제4、5조외주혈CD4+백분비[(44.1±5.8)%、(43.3±6.2)%]고우제1조[(33.5±3.1)%](P치균<0.05);제3、4、5조외주혈CD8+백분비[(21.3±3.3)%、(21.1±4.5)%、(20.5±4.1)%]저우제1조[(28.2±1.7)%](P치균<0.05).제4、5조검측미잡응효주입측폐조직연마혼장내γ-간우소수평위양성.결론 이건강일본장이토위모형국부면역자격가이인기전신면역응답반응.
Objective To observe local and systemic immunity effect of Mycobacterium vaccae gel administered in lung. Methods 60 Japanese long-eared rabbits were randomly divided into five groups.Group 1 was injected with 3 ml normal sodium. Group 2 was injected with 3 ml carbomer gel. Group 3,4,5 were injected with 3 ml carbomer gel with 3.75,7.50,9.37 mg/L Mycobacterium vaccae. The peripheral blood T cell subsets and interferon-γ were tested. Results Before sacrificed,CD3 percents in group 4 and group 5 [(55.8±9.7) % and (54.5±11.8) %] were higher than that in group 1 [(43.1±3. 0) %] (all P <0.05),CD4 + percents in group 4 and group 5 [ (44.1 ± 5.8) % and (43.3 ± 6.2) %] were higher than that ingroup 1 [(33.5± 3.1)%] (all P <0.05),CD8+ percents in group 3, 4, 5 [(21.3±3.3)%,(21.1 ± 4.5) % and (20. 5 ± 4.1) % ] were lower than that in group 1 [ (28.2 ±1.7) % ] (all P <0.05).Interferon-γ was positive in the lung tissues injected with carbomer gel with Mycobacterium vaccae in group 4 and group 5. Conclusions Local immune stimulation can cause systemic immune response in healthy Japanese long-eared rabbits.
