目的 比较结膜松弛症和正常人泪液中蛋白质成分的差异,从而探讨结膜松弛症的发病机制.方法 病例对照研究.收集结膜松弛症组和正常对照组各8例(8只眼),采用毛细管从每只眼收集15ul泪液,应用shotgun方法 检测泪液蛋白质,分别行溶液内酶解、反向高效液相色谱(RP-HPLC)分离、电喷雾离子化质谱(ESI-MS/MS)行质谱鉴定后生物信息分析.结果 结膜松弛症组泪液中共有蛋白质356个,正常人泪液中共有蛋白质352个,两组中有119个蛋白质相同.对泪液中蛋白质按基因本体注释(GOA)分类后,发现结膜松弛症组中出现的部分调节凋亡相关蛋白质、凋亡相关蛋白质及炎性反应相关蛋白质在正常对照组中缺失,并且在结膜松弛症泪液中发现了防御素.结论 shotgun方法 可有效分离和分析泪液蛋白质,结膜松弛症患者泪液中的特异性成分提示结膜松弛症可能与细胞凋亡和炎性反应有关.(中华眼科杂志,2009,45:135-140)
目的 比較結膜鬆弛癥和正常人淚液中蛋白質成分的差異,從而探討結膜鬆弛癥的髮病機製.方法 病例對照研究.收集結膜鬆弛癥組和正常對照組各8例(8隻眼),採用毛細管從每隻眼收集15ul淚液,應用shotgun方法 檢測淚液蛋白質,分彆行溶液內酶解、反嚮高效液相色譜(RP-HPLC)分離、電噴霧離子化質譜(ESI-MS/MS)行質譜鑒定後生物信息分析.結果 結膜鬆弛癥組淚液中共有蛋白質356箇,正常人淚液中共有蛋白質352箇,兩組中有119箇蛋白質相同.對淚液中蛋白質按基因本體註釋(GOA)分類後,髮現結膜鬆弛癥組中齣現的部分調節凋亡相關蛋白質、凋亡相關蛋白質及炎性反應相關蛋白質在正常對照組中缺失,併且在結膜鬆弛癥淚液中髮現瞭防禦素.結論 shotgun方法 可有效分離和分析淚液蛋白質,結膜鬆弛癥患者淚液中的特異性成分提示結膜鬆弛癥可能與細胞凋亡和炎性反應有關.(中華眼科雜誌,2009,45:135-140)
목적 비교결막송이증화정상인루액중단백질성분적차이,종이탐토결막송이증적발병궤제.방법 병례대조연구.수집결막송이증조화정상대조조각8례(8지안),채용모세관종매지안수집15ul루액,응용shotgun방법 검측루액단백질,분별행용액내매해、반향고효액상색보(RP-HPLC)분리、전분무리자화질보(ESI-MS/MS)행질보감정후생물신식분석.결과 결막송이증조루액중공유단백질356개,정상인루액중공유단백질352개,량조중유119개단백질상동.대루액중단백질안기인본체주석(GOA)분류후,발현결막송이증조중출현적부분조절조망상관단백질、조망상관단백질급염성반응상관단백질재정상대조조중결실,병차재결막송이증루액중발현료방어소.결론 shotgun방법 가유효분리화분석루액단백질,결막송이증환자루액중적특이성성분제시결막송이증가능여세포조망화염성반응유관.(중화안과잡지,2009,45:135-140)
Objective To analysis tear proteins by using shotgun strategy, and to study pathogenesis mechanism of conjunctivochalasis by comparing tear proteins between the conjunctivochalasis and normal body. Methods Tears were obtained from 8 conjunctivochalasisi cases and 8 normal controls. Fifteen microliters of tears was collected by microcapillary tubes from each eye. Shotgun strategy was used for tear protein analysis. Trypsin digestion in-solution, separation of peptide mixture by reverse-phase high-pressure liquid chromatograph (RP-HPLC) followed by electrosprayionization mass spectrometry (ESI-MS/ MS) identification and bioinformation analysis were used in this study. Results Three hundred and fifty-six proteins were exised in conjunctivochalasis patients and 352 proteins in the normal controls, and among them 119 proteins were the same. The functias of tear proteins were classified with GOA, which found some apoptosis regulation proteins and apoptosis related proteins and inflammatory response related proteins in conjuntivochalasis but not in normal controls group. Defensin was also found in conjuntivochalasis. Conclusion Shotgun strategy can separate and anlysis tear proteins effectively, which provide a new method for tear protein component of conjunctivochalasis. The special component in conjunctivochalasis tear show conjunctivochalasis maybe related to cell apoptosis and inflammatory. (Chin J Ophthalmol, 2009,45:135-140)
