目的 探讨二次膜分离提取藏牦牛血凝血酶的工艺.方法 根据藏牦牛血凝血酶原及其凝血酶的分子量,进行二次膜分离技术提取藏牦牛血凝血酶.结果 采用二次膜分离技术提取藏牦牛血凝血酶.其比活力与活力回收分别比有机溶剂沉淀法提高了76.72 U/mg蛋白质和35.0%;其比活力比一次膜分离法提高了41.14 U/mg蛋白质.结论 工艺可靠,方法可行,适宜于中小企业的生产.

Objective To establish crafts to extract thrombin from the blood of the Tibet yak by double-membrane separation. Methods According to molecular weights of blood thrombino- gen and thrombin of Tibetan yak,thrombin from the blood of the Tibet yak was extracted by double-membrane separation. Results The specific activity and activity recollection of thrombin were improved by 76.72 U/mgpro and 35.0% respectively when compared with organic solvent precipitation method,and the specific activity of thrombin was improved by 41.14 U/mg pro when compared with single membrane separation. Conclusion The craft is credible and feasible,the and can be used for the production of small and medium-sized scales.
