家鸡生长激素释放激素(GHRH)的成热肽段区存在一个酰胺化位点,提示具27个氨基酸残基的羧基端酰胺化多肽(cGHRH1-27NH2)可作为家鸡生长激素释放激素受体(GHRHR)的配体.为验证该假说,利用pGL3-CRE报告基因系统,在培养的中国仓鼠卵巢细胞中,探究人工合成cGHRH1-27N2多肽对家鸡两个GHRH受体的激活潜能.结果显示:cGHRH1-27NH2可高效激活家鸡两个GHRH受体,并且该多肽对GHRH一型受体(cGHRHR1:EC50:0.18 nmol/L)的激活效能略高于其对GHRH二型受体(cGHRHR2:EC5o:0.32 nmol/L)的激活效能.这些研究结果清楚表明GHRH1-27NH2可作为家鸡两个GHRH受体的配体.

Previous study revealed that there exists an amidation site within the putative mature chicken growth hormone-releasing hormone (GHRH) peptide region,pointing to a possibility that GHRH peptide of 27 amino acids with an amidated C-terminus (cGHRH1-27NH2) is a ligand for chicken GHRH receptors (GHRHRs).To substantiate this hypothesis,using a pGL3-CRE reporter system,the poten cies of cGHRH1-27NH2 in activating the two GHRH receptors (namely GHRHR1 and GHRHR2) were examined in cultured Chinese hamster ovary (CHO) cells in this study.Results showed that GHRH1-27NH2 could activate the two GHRH receptors potently and cGHRH1-27NH2 is slightly more potent in activating cGHRHR1 (EC50:0.18 nmol/L) than in activating cGHRHR2 (EC50:0.32 nmol/L).The high potencies of cGHRH1-27NH2 in activating cGHRHRs clearly indicate that cGHRH1-27NHZ is a ligand for the two GHRHRs in chickens.
