目的研究体外小剂量化疗药物尼莫司汀(ACNU)、顺铂(CDDP)、卡铂(cap)(均抗癌药)持续给药对人胶质母细胞瘤株U87的生长抑制作用.方法不同浓度的ACNU、CDDP、CBP分别作用于U87,24 h后撤药(A组)和持续作用于U87 72 h(B组),用MTT法在24、48、72 h分别检测药物的2种作用方式对U87细胞的生长抑制率.结果不同浓度ACNU的2种方式,作用48、72 h对U87的生长抑制率差异无显著性(P>0.05);不同浓度CDDP和CBP持续作用48、72 h(B组)对U87的生长抑制率显著高于24 h撤药组(A组),2组比较差异性显著(P<005).结论小剂量化疗药物在一定的浓度范围内持续作用,对U87的体外生长抑制作用与药物浓度和作用时间成正比.

Objective To investigate the continous inhibitory effect of low dose nimustine (ACNU) , cisplatin(CDDP) and carboplatin(CBP) on the growth of glioblastoma cell line U87 in vitro. Methods U87 cells are divided into 2 groups: One mode is 24 h incubation of U87 with different concentration of ACNU, CDDP and CBP. DMEM was discarded, and incubate for another 24 h or 48 h in fresh DMEM without drugs ( A group). Another mode is U87 incubated with ACNU, CDDP and CBP respectively. The growth inhibition rate of U87 was detected respectively by MTT assay in 24 h, 48 h and 72 h (B group). Results The inhibitory rate of ACNU on the growth of U87 by 2 incubation modes of 48 h and 72 h had no difference (P >0. 05). The inhibitory rate of CDDP and CBP on the growth of U87 by continous incubation group ( B group) is much higher than the group with discard drugs after 24 h (A group) after 48 h and 72 h incubation. The difference of the 2 groups is prominent (P <0.05). Conclusion The inhibitory rate on the growth of U87 in vitro is proportional to the concentration and time of low dose chemotherapy drugs.
