[目的]为了从成熟石榴叶片中提取高质量、高产量的基因组DNA.[方法]针对成熟石榴叶片含多糖、多酚的特性,用改良CTABⅠ及改良CTAB Ⅱ法分别提取2个品种(墨石榴和青皮软籽石榴)的成熟石榴鲜叶、冻叶、干叶基因组DNA,并通过琼脂糖凝胶电泳、紫外分光光度计、酶切鉴定.[结果]改良CTAB Ⅱ法提取的基因组DNA电泳时点样孔干净,无拖带,OD_(260 nm)/OD_(280 nm)为1.7~1.9,酶切完全,其质量、产量都高于改良CTAB Ⅰ法.[结论]改良CTAB Ⅱ法是提取成熟石榴叶片中高质量、高产量基因组DNA的有效方法.

[Objective] The research aimed to extract the high quality and yield genomic DNA from mature pomegranate (Punica granatum Linn. ) leaves. [ Method ] According to the character of containing amylase and polyphenol, genomic ONA in fresh leaves, frozen leaves and dry leaves of 2 kinds of mature pomegranates were extracted by improved CTAB I and CTAB U method. And they were identified by agarose gel elec-trophoresis,ultraviolet spectrophotometer method and enzyme digestion. [Result] The results showed that the sample hole was clean and no pulling, OD_(260nm)/OD_(280nm) was 1.7-1.9 when the genomic DNA extracted by improved CTAB II method was electrophoresis. And its quality and yield was high than that extracted by improved CTAB I method. [ Conclusion] The improved CTAB H method was the effective method of extracting the high quality and yield genomic DNA from mature pomegranate (Punica granatum Linn. ) leaves.
