本文首先对铜配合物催化DNA氧化断裂的机制及相关因素,包括DNA的结合能力与方式、活性氧物种的形成和活性氧物种对底物的损伤等进行了讨论,然后结合本组的工作对配合物的结构及其对DNA切割性能的影响分别进行了总结,探讨了核数、核种类、位阻、电荷、结合方式以及氧化还原电位等因素的影响,同时还对光激发铜配合物断裂DNA的机制和性能进行了分析,最后对特异性含铜DNA断裂试剂的不同设计策略及相关切割行为进行了系统总结,这不仅有利于对铜配合物促进DNA断裂行为的系统理解,而且对于进一步设计高效含铜人工核酸酶以实现DNA的特异性断裂具有良好的指导意义.
本文首先對銅配閤物催化DNA氧化斷裂的機製及相關因素,包括DNA的結閤能力與方式、活性氧物種的形成和活性氧物種對底物的損傷等進行瞭討論,然後結閤本組的工作對配閤物的結構及其對DNA切割性能的影響分彆進行瞭總結,探討瞭覈數、覈種類、位阻、電荷、結閤方式以及氧化還原電位等因素的影響,同時還對光激髮銅配閤物斷裂DNA的機製和性能進行瞭分析,最後對特異性含銅DNA斷裂試劑的不同設計策略及相關切割行為進行瞭繫統總結,這不僅有利于對銅配閤物促進DNA斷裂行為的繫統理解,而且對于進一步設計高效含銅人工覈痠酶以實現DNA的特異性斷裂具有良好的指導意義.

The oxidative DNA cleavage promoted by copper complexes and the related factors including the DNA binding mode/ability, the formation of reactive oxygen species (ROS) and the ROS damaging DNA were initially discussed in this review. Then, the copper complex structral factors affected the DNA cleavage, such as the metal center, nuclear number, steric hinderance, charge number, DNA binding mode and redox potential of copper center were surveyed based on the reported works of ours and others. In addition, the photo-induced DNA cleavage behavior of copper complexes was also investigated. Finally, the different strategies of copper nuclease construction for the specific DNA cleavage were systematically discussed. All these should not only favor to understand the oxidative DNA cleavage mediated by copper complexes but also promote to construct novel copper-containing artificical nucleases in the future.
