用差速离心法和改进的碱裂解法分离纯化遵义小钝吻鸢毛线粒体DNA,紫外检测其OD260/OD280在1.78-1.85,并计算出该鱼肝组织线粒体DNA的含量为0.613μg/g。纯化样品经紫外分析仪在200-290nm波长范围内扫描,呈现典型的DNA吸收峰,其最大吸收峰在259~260nm。以hDNA/HindIII的完全酶切片段作为相对分子量标准,利用电泳迁移率与分子量的对数的线性关系,由已知片段大小推算出遵义小钝吻鸵mtDNA分子大小为(15.44±0.16)kb。

The mitochondrial DNA(mtDNA)from liver ofLeioxassis~ticrocrassirostris ZunyiensisXiao sp.nov.was isolated and purified by means of differential centrifugation and the improved alkaline lysisoThe OD260/OD280 was 1.78-1.85 by UV spectrophotometer,and the quality of mtDNA obtained was 0.613 μg/goUnder the wavelength scan of 200-290 nm by UV solution ,the Purification of DNA of the sample presented characteristic DNA absorption peaks and the maximum one in 259-260 nmoUsekDNA/Hind m restriction fragments as a relatively molecular weight standard, according to the electrephoretic migration rate was linearly related to the molecular weight, the mtDNA size was (15.44±0.16) kb in the fish .
