基于过氧化苯甲酰((C6H5CO)2O2)的强氧化性对多壁碳纳米管(multi-wall nanotubes,MWNTs)末端的切割效应,得到水溶性MWNTs',同时,利用聚丙烯酰胺(Polyacrylamide,PA)对MWNTs'室温磷光(room temperature phosphorescence,RTP)的增敏效应,开发了水溶性PA-MWNTs'磷光标记试剂.以MWNTs'-PA标记人绒毛膜促性腺激素-β-C-末端肽单克隆抗体(human chorionic gonadotrophin-antibody,AbHCG),获得可保持MWNTs'的RTP优良特性AbHCG-MWNTs'-PA标记产物.该AbHCG-MWNTs'-PA不仅能与人绒毛膜促性腺激素(human chorionic gonadotrophin,HCG)发生定量的特异性免疫反应,而且免疫反应产物HCG-AbHCG-MWNTs'-PA能发射更强的RTP信号,据此建立了固体基质室温磷光免疫法(solid substrate room temperature phosphorescence immunoassay,SSRTPIA)测定HCG的新方法.SSRTPIA的线性范围为0.040-20.0 pg HCG spot 1(样品体积0.40μL spot 1,对应浓度0.10-50.0 ng mL 1),检出限为0.017 pg spot 1(447/613 nm,双抗夹心法).该方法灵敏、准确、精密度好,已用于人血清中HCG含量的测定及人体疾病的诊断.

Based on the cutting effect on the end of multi-wall carbon nanotubes(MWNTs) by taking advantages of the strong oxidation of benzoyl peroxide((C6H5CO)2O2),the water-soluble multi-wall nanotubes(MWNTs') were obtained.At the same time,we developed a new phosphorescent labeling reagent MWNTs'-polyacrylamide(PA) utilizing the spiking effect of PA on the room temperature phosphorescence(RTP) of MWNTs'.MWNTs'-PA was used to label human chorionic gonadotropin-β-C-terminal peptide monoclonal antibody(AbHCG) to obtain labeling product AbHCG-MWNTs'-PA which maintained good characteristics of RTP of the MWNTs'.AbHCG-MWNTs'-PA could take specific immunoreaction with human chorionic gonadotropin(HCG) to form HCG-AbHCG-MWNTs'-PA,resulting in the increase of MWNTs' RTP signal.Thus,a new solid substrate room temperature phosphorescence immunoassay(SSRTPIA) for the determination of HCG was established.The linear range of SSRTPIA was 0.040-20.0 pg HCG spot-1(sample volume: 0.40 μL spot-1,corresponding concentration: 0.10-50.0 ng mL 1),the detection limit(LD) was 0.017 pg spot 1 for sandwich way at 447/613 nm(λexmax/λemmax).This sensitive,accurate and precise method was used for the determination of HCG and the diagnosis of human diseases.
