目的:观察清肝解郁方对老年性痴呆小鼠学习记忆功能及神经信使分子NO代谢的影响.方法:从21月龄昆明种小鼠筛选出老年性痴呆模型鼠,随机分为4组,即空白对照组,西药组,清肝解郁方小剂量、大剂量组,另设正常老年组,连续给药60天,用跳台实验检测学习记忆成绩;NBT组织化学法检测大脑NOS1活性.结果:清肝解郁方能显著改善老年性痴呆小鼠的学习记忆障碍,并明显增强中枢大脑皮质和海马NOS1活性及其纤维密度,其作用呈现出一定的量效关系.此外,研究还发现颞叶皮质、海马NOS1活性与学习记忆成绩呈显著性正相关.结论:清肝解郁方可明显改善老年性痴呆模型的学习记忆障碍,这一作用可能与增强其大脑皮质和海马NOS1活性,调节中枢神经信使分子NO代谢有关.

In order to observe the effects of purging fire and resuscitating prescription on the cognitive ability and metabolism of nerve messenger molecule NO in spontaneous aged dementia mice.Meothods: Spontaneous aged dementia mice were screened out from 21 months old Kunming mice and then were diveded randomly into 4 groups:control, western medicine, low and high dosage purging fire and resuscitating prescrigtion groups,in addition, normal aged mice group was also set, each group were given corresponding medicines, all groups were treated as a aforementioned for 60 days, using step-down test to estimate the ability of learning and memory.Brain tissue was made to frozen section,Using NBT histochemical technique to show the activity of NOS1 and using automatic micro-image analysis system to determine quantitatively.Results: Purging fire and resuscitating prescription could improve the disturbance of learning and memory in spontaneous aged dementia mice remarkably, moreover, could enhance the activity and fiber density of NOS1 in cerebra cortex and hippocampus.All these effects had a dose-response relationship, furthermore, the results showed the activity of NOS1 in temporal lobe cortex and hippocampus had close positive correlation with the ability of learning and memory.Conclusion: Activity of NOS1 in CNS had close correlation with the ability of learning and memory.Purging fire and resuscitating proscription could improve the disturbance of learning and memory in spontaneous aged dementia mice significantly,this effect might relate to its action of enhancing the activity of NOS1 in cerebra cortex and hippocampus,regulating the metabolism of nerve messenger molecule NO.
