以小麦T型细胞质雄性不育系75-369A和相应保持系75-369B为材料,用限制性内切酶BamH、EcoR、Hind完全酶解,以Oenothera mtDNA atp6,小麦线粒体基因nad3/rps12、cox1为探针进行Southern杂交。杂交结果表明,75-369A和75-369B在这3个基因上或附近有显著的组织结构差异,推测这些差异可能影响了线粒体基因组的正常功能,最终引起了75-369A雄性不育。
以小麥T型細胞質雄性不育繫75-369A和相應保持繫75-369B為材料,用限製性內切酶BamH、EcoR、Hind完全酶解,以Oenothera mtDNA atp6,小麥線粒體基因nad3/rps12、cox1為探針進行Southern雜交。雜交結果錶明,75-369A和75-369B在這3箇基因上或附近有顯著的組織結構差異,推測這些差異可能影響瞭線粒體基因組的正常功能,最終引起瞭75-369A雄性不育。

mtDNAs of T type wheat cytoplasmic male sterile lines 75-369A and their maintainers 75-369B were isolated and digested completely with restriction endonuclease BamH, EcoR, Hind. The mitochondrial genomic difference between CMS line and maintainers were revealed by Southern hybridization with probes of atp6 fromOenotheraandnad3/rps12,cox from wheat mitochondria. Southern hybridization showed the differences between these two lines. Discussion was made on the change of the DNA composition and the male sterile cytoplasm.
