探讨X射线诱导EL-4细胞G1期阻滞及相关蛋白的表达。彩PI荧光标记,流式细胞术检测细胞周期的变化。用单克隆抗体免疫荧光标记,流式细胞术检测蛋白表达的变化。结果表明,2.0Gy和4.0Gy照射后12-72h,G1期EL-4细胞百分数显著高于假照射组(p<0.05-p<0.001)。4.0Gy照射后EL-4细胞p53蛋白表达从照射后2h开始明显增高,持续至照射后24h(p<0.05-p<0.001);p21蛋白表达在照射后2h开始明显增高,持续至照射后48h(p<0.05-p<0.001);GADD45蛋白表达在照射后2h开始明显增高,持续至照射后24h(p<0.05-p<0.001);MDM2蛋白表达在照射后4h开始明显增高,持续至照射后24h(p<0.05-p<0.001)。结果提示,中等剂量X射线照射可诱导EL-4细胞G1期阻滞。p53、p21和GADD45蛋白表达在电离辐射诱导EL-4细胞G1期阻滞中起重要作用
探討X射線誘導EL-4細胞G1期阻滯及相關蛋白的錶達。綵PI熒光標記,流式細胞術檢測細胞週期的變化。用單剋隆抗體免疫熒光標記,流式細胞術檢測蛋白錶達的變化。結果錶明,2.0Gy和4.0Gy照射後12-72h,G1期EL-4細胞百分數顯著高于假照射組(p<0.05-p<0.001)。4.0Gy照射後EL-4細胞p53蛋白錶達從照射後2h開始明顯增高,持續至照射後24h(p<0.05-p<0.001);p21蛋白錶達在照射後2h開始明顯增高,持續至照射後48h(p<0.05-p<0.001);GADD45蛋白錶達在照射後2h開始明顯增高,持續至照射後24h(p<0.05-p<0.001);MDM2蛋白錶達在照射後4h開始明顯增高,持續至照射後24h(p<0.05-p<0.001)。結果提示,中等劑量X射線照射可誘導EL-4細胞G1期阻滯。p53、p21和GADD45蛋白錶達在電離輻射誘導EL-4細胞G1期阻滯中起重要作用

To investigate G1 arrest and the expressions of relative proteins in EL-4 eclls induced by ionizing radiation,flow cytometry(FCM)was used to analyze cell cyle througyh staining eclls with propidium iodide(PI).And FCM was also employed to analyze protein expression of EL-4 cells stained with monoclonal and polyclonal antibodiss\ for their immunofluorescence.It was found that the number of EL-4 cells in G1 phase increased significantly 12-72h after X-irradiation with doses of 2.0 Gy and 4.0Gy.The results also showed significant increases of p53 protein expression at 2h to 24h(p<0.05-p<0.001),p21 protein expression at 2h to 48h(p<0.05-p<0.001),GADD45 protein expression at 2h to 48h(p<0.05-p<0.001),and MDM2 ptroein expression at 4g to 24h(p<0.05-p<0.001) after 4.0Gy X-irradiation.These results suggest that G1 arrest of EL-4 cells could be induced by ionizing radiation and the expression of p53,p21and GADD45 proteins may play an importatnt role in the mechanism.
