目的了解人转铁蛋白(Tf)与甲氨喋呤(MTX)结合物(Tf-MTX)对人肝细胞和人肝癌细胞体外细胞毒的特点。方法采用氧化葡聚糖(Dex)T-40作为中介体,联结Tf与MTX制备人转铁蛋白-甲氨喋呤结合物(Tf-MTX),用四甲偶氮唑蓝法比较结合物对人肝细胞L-02及人肝癌细胞Bel 7404的体外杀伤强度。结果体外细胞毒试验显示,Tf-MTX对人肝细胞L-02作用48 h的IC50是对人肝癌细胞Bel 7404的3.36倍,而MTX对上述细胞作用1 h的IC50是Tf-MTX的3.22倍。结论 Tf-MTX对肿瘤细胞Bel 7404的杀伤作用有选择性。

목적료해인전철단백(Tf)여갑안첩령(MTX)결합물(Tf-MTX)대인간세포화인간암세포체외세포독적특점。방법채용양화포취당(Dex)T-40작위중개체,련결Tf여MTX제비인전철단백-갑안첩령결합물(Tf-MTX),용사갑우담서람법비교결합물대인간세포L-02급인간암세포Bel 7404적체외살상강도。결과체외세포독시험현시,Tf-MTX대인간세포L-02작용48 h적IC50시대인간암세포Bel 7404적3.36배,이MTX대상술세포작용1 h적IC50시Tf-MTX적3.22배。결론 Tf-MTX대종류세포Bel 7404적살상작용유선택성。
Objective To research the cytotoxicity effects of a human transferrin(Tf)-methotrexatum(MTX) conjugate to human hepatocellular L-02 cells and human hepatocellular carcinoma Bel 7404 cells. Methods Tf was selected to conjugate with MTX via dextran T-40 as liner to construct Tf-MTX. Growth inhibition of the Tf-MTX and MTX in L-02 cells was compared with that of the drugs in Bel 7404 cells as evaluated by the MTT-assay. Results The IC50 of the Tf-MTX for 48 h was 3.36-fold higher in L-02 cells than in the Bel 7404 cells. The IC50 of Tf-MTX in the cell lines above for 1 h was 3.22 times lower than that of MTX. Conclusion Conjugate Tf-MTX has a selective cytotoxicity to Bel 7404 cells.
