目的 探讨刚地弓形虫感染对小鼠睾丸细胞DNA的损伤作用.方法 选用9~10周龄雄性BALB/c小鼠20只,随机分为4组,每组5只,分为正常对照组(CG)和3个染虫组(G1,G2,G3).采用腹腔注射法建立小鼠刚地弓形虫感染的动物模型,CG组每只注射PBS 0.2 ml;染虫G1,G2,G3组注射纯化的刚地弓形虫速殖子悬液,每只注射刚地弓形虫速殖子的量依次为2.5×10 3、5×10 3、1×10 4,注射体积均为0.2 ml,应用单细胞凝胶电泳检测睾丸细胞DNA的损伤. 结果 正常对照组和染虫组的尾距分别为10.94±7.57、40.37±6.25、69.76±3.97、79.16±6.36,olive尾距分别为10.57±6.72、31.39±4.59、48.66±4.60、53.87±6.55,G1-G3组与CG组比较,差异具有统计学意义(P<0.01).结论 弓形虫感染可导致小鼠睾丸生殖细胞DNA不同程度的损伤.
目的 探討剛地弓形蟲感染對小鼠睪汍細胞DNA的損傷作用.方法 選用9~10週齡雄性BALB/c小鼠20隻,隨機分為4組,每組5隻,分為正常對照組(CG)和3箇染蟲組(G1,G2,G3).採用腹腔註射法建立小鼠剛地弓形蟲感染的動物模型,CG組每隻註射PBS 0.2 ml;染蟲G1,G2,G3組註射純化的剛地弓形蟲速殖子懸液,每隻註射剛地弓形蟲速殖子的量依次為2.5×10 3、5×10 3、1×10 4,註射體積均為0.2 ml,應用單細胞凝膠電泳檢測睪汍細胞DNA的損傷. 結果 正常對照組和染蟲組的尾距分彆為10.94±7.57、40.37±6.25、69.76±3.97、79.16±6.36,olive尾距分彆為10.57±6.72、31.39±4.59、48.66±4.60、53.87±6.55,G1-G3組與CG組比較,差異具有統計學意義(P<0.01).結論 弓形蟲感染可導緻小鼠睪汍生殖細胞DNA不同程度的損傷.
목적 탐토강지궁형충감염대소서고환세포DNA적손상작용.방법 선용9~10주령웅성BALB/c소서20지,수궤분위4조,매조5지,분위정상대조조(CG)화3개염충조(G1,G2,G3).채용복강주사법건립소서강지궁형충감염적동물모형,CG조매지주사PBS 0.2 ml;염충G1,G2,G3조주사순화적강지궁형충속식자현액,매지주사강지궁형충속식자적량의차위2.5×10 3、5×10 3、1×10 4,주사체적균위0.2 ml,응용단세포응효전영검측고환세포DNA적손상. 결과 정상대조조화염충조적미거분별위10.94±7.57、40.37±6.25、69.76±3.97、79.16±6.36,olive미거분별위10.57±6.72、31.39±4.59、48.66±4.60、53.87±6.55,G1-G3조여CG조비교,차이구유통계학의의(P<0.01).결론 궁형충감염가도치소서고환생식세포DNA불동정도적손상.
Objective To explore the adverse effect of Toxoplasma gondii infection on DNA in mice spermatogenic cells. Method 20 BALB/c male mice(9-10 weeks old)were randomized into 4 groups,5mice per group:one negative control group(CG),three experimental group(G1:2.5 × 10 3、G2:5×10 3 and G3:1 × 10 4 tachyzoites).The CG group was injected intraperitoneally with PBS and the experimental groups were inoculated intraperitoneally with T. gondii tachyzoites(RH strain).All mice were sacrificed 6 davs after infection and the testicular cells were applied into single ceil gel electrophoresis for detection of DNA damage.Result The tail moment mean value of CG was 10.94±7.57.The mean value of tail moments of three experimental groups were 40.37±6.25,69.76±3.97 and 79.16±6.36 respectively.While the olive tail moments of CG and G1,G2,G3 were 10.57±6.72,31.39±4.59,48.66±4.60,53.87±6.55 respectivelv.Significant differences in tail moment and olive tail moment were seen between the experimental groups and the negative control group(P<0.01). Conclusion T. gondii could cause DNA damage of mice spermatogenic cells.
