从盾叶薯蓣根状茎中分离并鉴定了9株内生真菌,经悬浮培养14 d,分别制备灭活菌丝和菌液浓缩物.其中,内生尖孢镰刀菌Dzf17能有效地提高盾叶薯蓣无菌苗和培养细胞薯蓣皂苷元的含量和产率,且灭活菌丝的诱导效果要强于菌液浓缩物.Dzf17灭活菌丝处理无菌苗,薯蓣皂苷元的产率为78.697 mg/L,是对照(27.471 mg/L)的2.865倍;用Dzf17菌液浓缩物处理无菌苗,皂苷元产率为41.822 mg/L,是对照的1.522倍.Dzf17灭活菌丝处理培养细胞,薯蓣皂苷元的产率为1.391 mg/L,是对照(0.691 mg/L)的2.013倍;用Dzf17菌液浓缩物处理培养细胞,皂苷元产率为1.214 mg/L,是对照的1.757倍.结果表明,在盾叶薯蓣无菌苗或细胞培养中添加一定量的内生真菌灭活菌丝或菌液浓缩物对于提高薯蓣皂苷元含量和产量将是一种有效的方法.

Nine endophytic fungal isolates were separated from the fresh healthy rhizomes of Dioscorea zingiberensis, and they were taxonomically identified by both morphological characters and r-DNA molecular analysis.Their autoclaved my-celia and concentrated filtrates were prepared after two weeks of suspension culture.Of them,isolate Dzf17 ,which was i-dentified as Fusarium oxysporttm, was found to effectively increase diosgenin content and yield of the seedlings and sus-pension cells of D.zingiberensis,especially the elicitation effect of the autoclaved mycelia was stronger than that of the concentrated filtrate.Under the treatment of autoclaved Dzf17 mycelia on the seedlings or culture cells, the diesgenin yield of the seedlings was 78.697 mg/L that was 2.865 fold of the control (27.471 mg/L), and the diesgenin yield of the culture cells was 1.391 mg/L that was 2.013 fold of the control (0.691 mg/L).Similarly,under the treatment of Dzf17 concentrated filtrate, the diesgenin yield of the seedlings was 41.822 mg/L that was 1.522 fold of the control, and the diosgenin yield of the culture cells was 1.214 mg/L that was 1.757 fold of the control.The results indicated that ad-dition of the autoclaved mycelia or concentrated filtrates from the endophytic fungi should be an effective way to increase diosgenin content and yield in cell suspension and seedling culture.
