目的 观察丁酸钠对大鼠小肠上皮细胞株-6(intestinal epithelium cell line No.6,IEC-6)细胞增殖和凋亡的作用. 方法 不同浓度丁酸钠(低浓度2 mmol/L,中浓度8 mmol/L,高浓度14、20 mmol/L)处理IEC-6细胞,细胞计数试剂盒CCK-8法检测细胞增殖情况,分光光度法检测细胞内Caspase-3活性程度,流式细胞术检测细胞凋亡率. 结果 (1)加入丁酸钠(2、8、14、20mmol/L)共培养24 h后,2、8 mmol/L丁酸钠组细胞增殖速度比对照组快,差异有统计学意义(P<0.01);48 h后,14、20 mmol/L丁酸钠组细胞增殖较对照组明显受抑制,差异有统计学意义(P<0.01);72 h后,8、14、20 mmol/L丁酸钠组增殖亦明显受抑制,与对照组比较差异有统计学意义(P<0.01).(2)加入丁酸钠(2、8、14 mmol/L)共培养12 h后,8、14 mmol/L丁酸钠组与对照组比较Caspase-3活性明显升高,差异有统计学意义(P<0.05);24 h后,2、8、14 mmol/L丁酸钠组与对照组比较Caspase-3活性均升高,差异有统计学意义(P<0.05);48 h后,8、14 mmol/L丁酸钠组与对照组比较Caspase-3活性亦升高,差异有统计学意义(P<0.05).(3)加入丁酸钠(2、8、14 mmol/L)共培养12、24、48 h后,8、14 mmol/L丁酸钠组与对照组比较IEC-6细胞凋亡率均有升高,差异有统计学意义(P<0.05). 结论 中、高浓度丁酸钠有诱导IEC-6细胞凋亡,抑制IEC-6细胞增殖作用,而低浓度丁酸钠没有这种作用.

목적 관찰정산납대대서소장상피세포주-6(intestinal epithelium cell line No.6,IEC-6)세포증식화조망적작용. 방법 불동농도정산납(저농도2 mmol/L,중농도8 mmol/L,고농도14、20 mmol/L)처리IEC-6세포,세포계수시제합CCK-8법검측세포증식정황,분광광도법검측세포내Caspase-3활성정도,류식세포술검측세포조망솔. 결과 (1)가입정산납(2、8、14、20mmol/L)공배양24 h후,2、8 mmol/L정산납조세포증식속도비대조조쾌,차이유통계학의의(P<0.01);48 h후,14、20 mmol/L정산납조세포증식교대조조명현수억제,차이유통계학의의(P<0.01);72 h후,8、14、20 mmol/L정산납조증식역명현수억제,여대조조비교차이유통계학의의(P<0.01).(2)가입정산납(2、8、14 mmol/L)공배양12 h후,8、14 mmol/L정산납조여대조조비교Caspase-3활성명현승고,차이유통계학의의(P<0.05);24 h후,2、8、14 mmol/L정산납조여대조조비교Caspase-3활성균승고,차이유통계학의의(P<0.05);48 h후,8、14 mmol/L정산납조여대조조비교Caspase-3활성역승고,차이유통계학의의(P<0.05).(3)가입정산납(2、8、14 mmol/L)공배양12、24、48 h후,8、14 mmol/L정산납조여대조조비교IEC-6세포조망솔균유승고,차이유통계학의의(P<0.05). 결론 중、고농도정산납유유도IEC-6세포조망,억제IEC-6세포증식작용,이저농도정산납몰유저충작용.
Objective To investigate the effects of sodium butyrate on the proliferation and apoptosis of intestinal epithelium eell line No.6(IEC-6)of the rats. Methods IEC-6 cells were treated with sodium butyrate at different concentrations(low concentration 2 mmol/L,medium 8mmol/L,high 14 and 20 mmol/L)or incubated in medium alone as a control.Morphological changes of IEC-6 cells were observed by inverted microscope,and cell proliferation was assessed by cell counting kit-8(CCK-8)assay.Caspase-3 activity was measured and flow cytometry was used to assay the apoptosis. Results (1)The growth rates of the cells treated with sodium butyrate in concentrations of 2,8 mmol/L for 24 h were higher than those of control group(P<0.01);At 48 h,the 14,20 mmol/L sodium butyrate groups had the lower growth rates than control group(P<0.01);At 72 h,the 8,14,20 mmol/L sodium butyrate groups had the lower growth rates than control group(P<0.01).(2)The caspase-3 activity from cell lysates of IEC-6 cells treated with sodium butyrate in concentrations of 8,14 mmol/L for 12 h were higher than that of control group(P<0.05);At 24 h,the 2,8,14 mmol/L sodium butyrate groups had the higher caspase-3 activity than control group(P<0.05);At 48 h,the 8,14 mmol/L sodium butyrate groups had the higher caspase-3 activity than eontrol group(P<0.05).(3)The rates of cell apoptosis in groups treated with sodium butyrate in concentrations of 8,14 mmol/L for 12,24,48 h were higher than those of control group intestinal epithelial cells,and the effects are dose and time dependent.
