对经诱变后黑曲霉固体发酵生产酸性蛋白酶的培养基组成及发酵条件进行优化.结果表明,该菌发酵产酶的最适培养基和条件为米糠20 g,碳源玉米粉60g/kg,氮源硝酸铵5g/kg,MnCl2 4 g/kg,料液比1.0:1.5,调初始pH值7,以6mL/100g的接种量接种,在40℃、180 r/min下发酵72 h,摇瓶产酶达到1120.0 U/mL,比基础培养基提高了近2.9倍.
對經誘變後黑麯黴固體髮酵生產痠性蛋白酶的培養基組成及髮酵條件進行優化.結果錶明,該菌髮酵產酶的最適培養基和條件為米糠20 g,碳源玉米粉60g/kg,氮源硝痠銨5g/kg,MnCl2 4 g/kg,料液比1.0:1.5,調初始pH值7,以6mL/100g的接種量接種,在40℃、180 r/min下髮酵72 h,搖瓶產酶達到1120.0 U/mL,比基礎培養基提高瞭近2.9倍.
대경유변후흑곡매고체발효생산산성단백매적배양기조성급발효조건진행우화.결과표명,해균발효산매적최괄배양기화조건위미강20 g,탄원옥미분60g/kg,담원초산안5g/kg,MnCl2 4 g/kg,료액비1.0:1.5,조초시pH치7,이6mL/100g적접충량접충,재40℃、180 r/min하발효72 h,요병산매체도1120.0 U/mL,비기출배양기제고료근2.9배.
The medium composition and fermentation conditions were optimized for solid-state fermentation of Aspergillus niger.The experimental results showed that the optimum medium and conditions were as follows:rice bran 20g,6 g/kg Corn meal,ammonium nitrate nitrogen 5 g/kg,MnCl_2 4 g/kg,liquid ratio 1.0:1.5,adjusted initial pH7,6 mL/100g of the volume of cornvaccine inoculation,40℃、180 r/min fermentation time 72 h,in shake flask enzyme production rate reached 1120.0 U/mL,2.9 times more than that under base medium.
