目的 探讨肿瘤坏死因子α诱导的蛋白8-2(tumor necrosis factor-induced protein 8 like 2,TIPE2)在慢性阻塞性肺疾病(COPD)患者外周血单个核细胞(PBMC)中的表达及其在COPD发病中的作用.方法 选择42例符合标准的COPD患者及37名健康对照者,用荧光定量PCR法检测其TIPE2的表达水平,用ELISA法检测血清中肿瘤坏死因子α(TNF-α)、白介素6(IL-6)的含量,由定量数据分析其直线相关关系.结果 COPD组TIPE2表达水平明显低于正常组(11.25±1.37比4.50±1.04,P＜0.01),而炎性细胞因子TNF-α、IL-6的含量明显高于正常组；COPD组TIPE2的表达水平与血清中TNF-α、IL-6的水平均呈负相关,而正常组二者之间无明显相关性.结论 COPD患者PBMC中TIPE2表达下降,且其表达水平与炎性细胞因子TNF-α、IL-6呈负相关,TIPE2可能在COPD的发病中发挥重要作用.

Objective To investigate the expression of tumor necrosis factor-induced protein 8 like 2 (TIPE2) in peripheral blood mononuclear cells (PBMC) from patients with chronic obstructive pulmonary disease (COPD) and access the role of TIPE2 in the pathogenesis of COPD.Methods The expression of TIPE2 was examined in PBMC from 42 COPD patients and 37 healthy controls by real time PCR.The concentrations of TNF-α and IL-6 in serum were determined by ELISA,and then their linear correlation was analyzed using quantitative datas.Results The TIPE2 expression was significantly down-regulated in COPD patients compared with healthy controls (11.25±1.37 vs 4.50± 1.04,P ＜0.01),while the levels of proinflammatory cytokines TNF-α and IL-6 in sera were higher in the COPD group compared with healthy controls.Furthermore,the TIPE2 expression levels negatively correlate with TNF-α and IL-6 in sera in the COPD group,while no significant correlations were observed between the TIPE2 expression levels and TNF-α and IL-6 in sera in healthy controls.Conclusions The expression levles of TIPE2 mRNA are decreased in PBMC from COPD patients compared with healthy controls and negatively correlated with TNF-α and IL-6 in sera,suggesting that TIPE2 may play an important role in the pathogenesis of COPD.
