目的:观察8-甲氧沙林对体外培养黑素细胞内游离钙浓度、细胞骨架蛋白形成的影响.方法:正常人黑素细胞取自包皮环切术切取的包皮,用激光共聚焦扫描显微镜检测细胞内游离钙浓度,用罗丹明结合毒伞素对细胞骨架蛋白进行特异性染色.结果:8-甲氧沙林可使细胞内游离钙离子浓度升高,促进黑素细胞骨架蛋白合成.结论:8-甲氧沙林可能通过诱导黑素细胞内钙离子浓度升高和细胞骨架actin蛋白生成,从而影响黑素细胞的迁移.
目的:觀察8-甲氧沙林對體外培養黑素細胞內遊離鈣濃度、細胞骨架蛋白形成的影響.方法:正常人黑素細胞取自包皮環切術切取的包皮,用激光共聚焦掃描顯微鏡檢測細胞內遊離鈣濃度,用囉丹明結閤毒傘素對細胞骨架蛋白進行特異性染色.結果:8-甲氧沙林可使細胞內遊離鈣離子濃度升高,促進黑素細胞骨架蛋白閤成.結論:8-甲氧沙林可能通過誘導黑素細胞內鈣離子濃度升高和細胞骨架actin蛋白生成,從而影響黑素細胞的遷移.
목적:관찰8-갑양사림대체외배양흑소세포내유리개농도、세포골가단백형성적영향.방법:정상인흑소세포취자포피배절술절취적포피,용격광공취초소묘현미경검측세포내유리개농도,용라단명결합독산소대세포골가단백진행특이성염색.결과:8-갑양사림가사세포내유리개리자농도승고,촉진흑소세포골가단백합성.결론:8-갑양사림가능통과유도흑소세포내개리자농도승고화세포골가actin단백생성,종이영향흑소세포적천이.
Objective: To investigate the effects of 8-methoxypsoralen on human melanocytes [Ca2+]i and cytoskeleton actin organization in vitro. Methods: Human melanocytes were obtained from normal foreskins.Laser confocal microscope was employed to measure [Ca2+]i and rhodamine-conjugated phalloidin was used to visualize the cytoskeleton actin. Results: 8-methoxypsoralen increased [Ca2+]i and induced organization of actin stress fiber cytoskeleton. Conclusion: 8-methoxypsoralen might influence the migration of melanocytes by increasing the intracellular free Ca2+ concentration and cytoskeleton actin reorganization.
