目的 通过研究淫羊藿苷(icariin,ICA)对骨髓间充质干细胞(bone mesenchymal stem cells,BMSCs)增殖和骨向分化能力的影响,并观察其促BMSCs骨向分化过程中对细胞因子转化生长因子β1(TGFβ1)、骨形成蛋白-2(BMP-2)表达的影响,分析可能的作用途径,为阐释其防治骨质疏松(osteoporosis,OP)的作用机理提供实验依据,并为以TGF-β1,、BMP-2作为筛选防治OP药物进行实验研究.方法 以碱性磷酸酶(ALP)等指标确定干预药物不同浓度梯度中最佳促BMSCs骨向分化剂量;分组实验,通过检测诱导分化过程中ALP和钙化结节等指标的表达,来观察其对BMSCs骨向分化能力的影响;采用ELISA法检测骨向分化过程中TGF-β1、BMP-2的表达.结果 (1)ICA最佳促BMSCs骨向分化浓度为1×10-9mol/L.ICA 10-9组能促进成骨指标的表达和TGF-β1和BMP-2分泌增加;(2)能促BMSCs骨向分化能力的ICA各诱导组均伴随TGF-β1和BMP-2分泌增加.结论 ICA能促进BMSCs骨向分化;在BMSCs骨向分化过程中,上调TGFβ1、BMP-2的表达量可能是各干预药物促进BMSCs骨向分化的作用机制之一.

목적 통과연구음양곽감(icariin,ICA)대골수간충질간세포(bone mesenchymal stem cells,BMSCs)증식화골향분화능력적영향,병관찰기촉BMSCs골향분화과정중대세포인자전화생장인자β1(TGFβ1)、골형성단백-2(BMP-2)표체적영향,분석가능적작용도경,위천석기방치골질소송(osteoporosis,OP)적작용궤리제공실험의거,병위이TGF-β1,、BMP-2작위사선방치OP약물진행실험연구.방법 이감성린산매(ALP)등지표학정간예약물불동농도제도중최가촉BMSCs골향분화제량;분조실험,통과검측유도분화과정중ALP화개화결절등지표적표체,래관찰기대BMSCs골향분화능력적영향;채용ELISA법검측골향분화과정중TGF-β1、BMP-2적표체.결과 (1)ICA최가촉BMSCs골향분화농도위1×10-9mol/L.ICA 10-9조능촉진성골지표적표체화TGF-β1화BMP-2분비증가;(2)능촉BMSCs골향분화능력적ICA각유도조균반수TGF-β1화BMP-2분비증가.결론 ICA능촉진BMSCs골향분화;재BMSCs골향분화과정중,상조TGFβ1、BMP-2적표체량가능시각간예약물촉진BMSCs골향분화적작용궤제지일.
Objective To research the effect of icariin(ICA)on the proliferation and differentiation of bone mesenchymal stem cells(BMSCs)and to study its influence on the expressions of transforming growth factor-β1(TGF-β1)and bone morphogenetic protein-2(BMP-2)in the progress of BMSCs differentiating into osteoblast,for providing an experimental evidence to explain the mechanism of ICA,also for exploring the feasibility of establishing a platform upon TGF-β1 and BMP-2 to screen out the medicine in preventing and treating osteoporosis.Methods After the most effective concentration of ICA for promoting the differentiation of BMSCs into osteoblast was judged with the indices like alkaline phosphatase(ALP),etc.,a grouped experiment was conducted for the sake of studying the effect and mechanism of ICA in its process of inducing BMSCs differentiation into osteoblast through detecting expression of ALP and calcium nodes,as well as the expressions of TGF-β1 and BMP2 with ELISA.Results The most effective concentration of the ICA on the BMSCs differentiation was judged as 1×10-9 mol/L,ICA of that concentration showed effects in enhancing the expressions of osteoblast-indices and increasing the secretion of TGF-β1 and BMP-2.Besides,the increase of TGF-β1 and BMP-2 was revealed in all the groups,in which ICA showed its influence visibly.Conclusion ICA could promote the differentiation of BMSCs into osteoblast; the up-regulation of TGF-β1 and BMP-2 expressions is possibly one of the action mechanisms of various interventional drugs in their differentiation promoting progress.
