分析了宁波地区幽门螺杆菌(Helicobacter pylori,Hp)对左氧氟沙星的耐药情况,探讨了Hp对左氧氟沙星的耐药机制.取患者胃黏膜组织分离培养Hp,检测Hp对左氧氟沙星的耐药率;提取左氧氟沙星耐药Hp DNA,PCR扩增并测序比较.结果表明:宁波地区Hp对左氧氟沙星的耐药率为22.5%,左氧氟沙星耐药Hp菌株gyrA基因91位密码子发生点突变.提示宁波地区Hp对左氧氟沙星已产生一定的耐药性;左氧氟沙星耐药Hp gyrA基因突变可能是本地区左氧氟沙星耐药的重要原因之一.

In this paper,we examine the antibiotic resistance of Helicobacter pylori(Hp) to levofloxacin in Ningbo,and investigate its possible molecular mechanism of resistance.To determine the resistance rate of Hp to levofloxacin,Hp is detached from mucous membrane of patients’ stomachs,and is subsequently cultured.The levofloxacin-resistant Hp DNA is then extracted,and the gyrA gene is amplified by PCR from resistance and susceptive strains.These PCR-products are sequenced,and the nucleotide sequences are analyzed.The results show that the resistance rate of Hp to levofloxacin is found to be 22.5%,and levofloxacin resistance strains of Hp possesses mutation in codon 91.The Hp strain in Ningbo has developed resistance to levofloxacin,and mutation of gyrA gene plays an important role in the possible mechanism of resistance to levofloxacin.
