目的:研究延衰中药补肾益精方对大鼠衰老过程(8~24月龄)中左心室肌球蛋白重链(α、βMHC)基因转录 mRNA表达量的影响,进一步探讨补肾益方的延衰机理。方法:大鼠喂饲到18月龄时随机分成老年组和老年服药组,中药用量是成人用量的30倍,服药6个月。以8月龄大鼠作为成年对照组。大鼠处死后,以常规放免法测定血清甲状腺素T3和T4水平。用RT-PCR方法扩增三组动物左心室α、βMHC 基因的有关转录产物,以GAPDH扩增片段做相对定量。结果:老年服药组与老年组相比,T4增加到1.24倍(P<0.001),α MHC mRNA增加到1.39倍(P<0.05),βMHC mRNA下调到74.32%(P<0.05)。结论:延衰中药对大鼠衰老增龄中α、βMHC基因表达的衰老变化有逆转作用。

목적:연구연쇠중약보신익정방대대서쇠로과정(8~24월령)중좌심실기구단백중련(α、βMHC)기인전록 mRNA표체량적영향,진일보탐토보신익방적연쇠궤리。방법:대서위사도18월령시수궤분성노년조화노년복약조,중약용량시성인용량적30배,복약6개월。이8월령대서작위성년대조조。대서처사후,이상규방면법측정혈청갑상선소T3화T4수평。용RT-PCR방법확증삼조동물좌심실α、βMHC 기인적유관전록산물,이GAPDH확증편단주상대정량。결과:노년복약조여노년조상비,T4증가도1.24배(P<0.001),α MHC mRNA증가도1.39배(P<0.05),βMHC mRNA하조도74.32%(P<0.05)。결론:연쇠중약대대서쇠로증령중α、βMHC기인표체적쇠로변화유역전작용。
Aim:To investigate the effect of BuShen Yi Jing decoction on rat left-ventricular myosin heavy chain(MHC)α and βgene expression change with aging. Methods:Wistar rats were randomly divided into three groups:untreated 8 and 24 months of aging,administration drug 24 months of aging. Rats were administration buShen Yi Jing decoction six months with 30 times adults human dose. Rats serum thyroxine(T4) and triiodothyronine(T3) levels using an radioimmunoassay(RIA) from clinic assay,RT-PCR method was used to evaluate α and βMHC mRNA expression. Results: T4 level was increased 1.24 times of senescent administration drug rats contrast to senility control rats (P<0.001),αMHC mRNA enhanced 1.3 times (P<0.05) and β MHC mRNA decreased 74.32%(P<0.05). Conclusion: Bushen Yijing decoction had reversed effect on α and βMHC gene expression change with aging.
