以烤烟品种Hicks和Speight G-28为材料,针对烟草RAPD分析中PCR过程的Taq酶浓度、Mg2+浓度、退火温度、循环数、Pyimer浓度、dNTP浓度等因素进行了研究。结果表明,体系中含0.8 U Taq酶,40mmol/L MgCl2,10μmol/L引物,3.0 mmo1/L dNTP,循环数为40,退火温度36℃效果较好。从而确立适合烤烟RAPD分析的PGR程序主要条件,为RAPD技术应用于烟草的品种鉴定与品种资源研究奠定了基础。

With tobacco variety Hicks and Speight CG-28 as experimental materials, taqase concentration, Mg2+ concentration,annealing temperature,number of amplified cycles,primer concentration and dNTP concenhation of RAPD analysis were studied. The results indicated that under the ccndition of 0.8 U taqase, 40 mmo1/L MgCl2, 10μmol/L primer, 3.0mmol/L dNTP, annealing temperature at 36 ℃C and 40 cycles, the effect was very good. PR APD pattem founded in this experiment layed good foundations for application of BAPD analysis in tobacco variety identification and gennplasm research.
