目的 观察理气活血法对萎缩性胃炎癌前病变大鼠胃黏膜组织基因表达谱的影响.方法 使用金属弹簧、灌服盐水溶液和热糊的综合方法建立大鼠萎缩性胃炎癌前病变模型,运用基因芯片技术,检测理气活血法干预后萎缩性胃炎癌前病变大鼠基因的差异表达变化,筛选出差异表达基因,并应用Real-time PCR对主要差异基因进行鉴定.结果 共筛选出差异表达基因14个,其中上调基因8条,下调基因6条,共同构成了理气活血法干预大鼠萎缩性胃炎癌前病变的差异表达基因谱.其中蛋白酪氨酸激酶2(ptk2)在自然恢复组中明显下调,经理气活血法干预后明显上调.结论 理气活血法可上调ptk2基因的表达,从而调节肌动蛋白细胞骨架的重构,促进损伤的胃黏膜修复和愈合,这可能是理气活血法治疗萎缩性胃炎癌前病变的机理之一.
目的 觀察理氣活血法對萎縮性胃炎癌前病變大鼠胃黏膜組織基因錶達譜的影響.方法 使用金屬彈簧、灌服鹽水溶液和熱糊的綜閤方法建立大鼠萎縮性胃炎癌前病變模型,運用基因芯片技術,檢測理氣活血法榦預後萎縮性胃炎癌前病變大鼠基因的差異錶達變化,篩選齣差異錶達基因,併應用Real-time PCR對主要差異基因進行鑒定.結果 共篩選齣差異錶達基因14箇,其中上調基因8條,下調基因6條,共同構成瞭理氣活血法榦預大鼠萎縮性胃炎癌前病變的差異錶達基因譜.其中蛋白酪氨痠激酶2(ptk2)在自然恢複組中明顯下調,經理氣活血法榦預後明顯上調.結論 理氣活血法可上調ptk2基因的錶達,從而調節肌動蛋白細胞骨架的重構,促進損傷的胃黏膜脩複和愈閤,這可能是理氣活血法治療萎縮性胃炎癌前病變的機理之一.
목적 관찰리기활혈법대위축성위염암전병변대서위점막조직기인표체보적영향.방법 사용금속탄황、관복염수용액화열호적종합방법건립대서위축성위염암전병변모형,운용기인심편기술,검측리기활혈법간예후위축성위염암전병변대서기인적차이표체변화,사선출차이표체기인,병응용Real-time PCR대주요차이기인진행감정.결과 공사선출차이표체기인14개,기중상조기인8조,하조기인6조,공동구성료리기활혈법간예대서위축성위염암전병변적차이표체기인보.기중단백락안산격매2(ptk2)재자연회복조중명현하조,경리기활혈법간예후명현상조.결론 리기활혈법가상조ptk2기인적표체,종이조절기동단백세포골가적중구,촉진손상적위점막수복화유합,저가능시리기활혈법치료위축성위염암전병변적궤리지일.
Objective To observe the influence of TCM therapy of regulating qi and activating blood circulation on the gene expression profile of gastric mucosa in rats with precancerosis of atrophic gastritis.Methods The rat model of preeancerosis of atrophic gastritis was established by the synthetic method of metal spring insertion and feeding of NaCI solution and heat paste.The changes of differential gene expressions in the rats were detected by using gene chip technology after the intervention of the TCM therapy.The differential expression genes were screened and then identified by applying RT-PCR.Results There were totally 14 differential expression genes screened,and among them 8 were upregulated genes and 6 wero down-regulated genes.All of them consisted of the differential expression gene profde after the intervention of the TCM therapy in rat with precancerosis of atrophic gastritis,among which ptk2 was regulated significantly downward in the spontaneous recovery group and regulated significantly upward after the intervention of the TCM therapy.Conclusion The TCM therapy of regulating qi and activating blood circulation can regulate upward the gene expression of ptk2 in order to regulate the remodeling ofactin cytoskeleton,and improve the repairing and healing of gastric mucosa.It is may be one of the mechanisms of the TCM therapy for treating precancerosis of atrophic gastritis.
