[目的]筛选出高效微生物絮凝剂产生菌并优化其培养条件.[方法]利用平板培养基和液体培养基筛选出絮凝活性高且稳定的菌株,通过计算絮凝率评价其发酵液的絮凝活性,最后通过单因素试验确定所选出菌株的最佳培养条件.[结果]分离出13株具有絮凝活性的菌株,菌株MC3的发酵液对高岭土悬浊液的絮凝率可达80.8%.培养72 h和84 h后菌株发酵液的絮凝率分别为80.8%和81.2%.以玉米粉和葡萄糖为碳源培养60 h后菌株发酵液的絮凝率分别为92.2%和87.8%,而葡萄糖的絮凝效果更稳定.pH值6时,培养60 h后菌株发酵液的絮凝率为81.0%.菌株MC3的最佳培养条件为:用葡萄糖替代查氏培养基中的蔗糖,培养液初始pH值6,接种量为10%,在该条件下培养60 h和72 h后菌株MC3发酵液的絮凝率分别为92.9%和92.0%.[结论]该研究为微生物絮凝剂的生产奠定了试验基础.

[Objective] The study aimed to screen out the efficient producing bacteria of microbial flocculant and optimize their culture condition. L Method] The strains with high and stable flocculating activity were screened out by using plate medium and liquid medium and the flocculating activities of their fermented liquid were evaluated through calculating their flocculating rates. Finally, the optimum culture conditions of the selected strain were ascertained through single factor experiments. [Result] The 13 strains with flocculating activity were screened out and the flocculating rate of fermented liquid from the strain MC3 on kaolin suspension could be up to 80. 8%. The flocculating rates of fermented liquid from the strains cultured for 72 h and 84 h were 80. 8% and 81. 2% resp. The flocculating rates of fermented liquid from the strains cultured with corn flour and glucose as carbon source for 60 h were 92. 2% and 87. 8% resp. and the flocculating effect of glucose was more stable. When the pH value was 6, the flocculating rate of fermented liquid from the strains cultured for 60 h was 81.0%. The optimum culture condition of the strain MC3 was as follow: the sucrose in Czapek' s medium was replaced with glucose,the initial pH value of the culture liquid was 6 and the inoculation amount was 10% . Under this condition ,the flocculating rates of fermented liquid from the strain MC3 cultured for 60 h and 72 h were 92.9% and 92.0% resp. [ Conclusion ] The study laid an experimental foundation for the production of microbial flocculant.
