以恩施地区五鹤续断的根尖为材料,研究了预处理时间和解离时间对其染色体制片的影响,并用此最佳条件进行了五鹤续断根尖染色体核型分析.结果表明,0.1%秋水仙素预处理2.5h、1mol/L盐酸解离30min的处理条件为最佳;五鹤续断染色体数目为18条,核型公式为2n=18=12m+4sm+2st,核型为2A型.

The roots of Wu-He Dipsactts in Enshi were used to study the effect of pre-treating time and the time of dissociation on the slide-preparing of the chromosomes.The results revealed that the pretreatment with 0.1%colchicines 2.5h and 1mol/L HCl at 60 temperature degree 30min preformed well in the sectioning.This technique was employed to determine the karyotype of Wu-He Dipsacus.The results showed that the number of chromosomes in Wu-He Dipsacus was 18 and the formula was 2n=18=12m+4sm+2st,and karyotype asymmetry belonged to 2A type.
