Integrin- linked kinase Chronic allograft nephropathy(CAN) Transforming growth factor- β1 (TGF- β1 )
目的:探讨移植肾组织中整合素连接激酶(ILK)及Ⅳ型胶原的表达,及与转化生长因子-β1(TGF-β1)表达和慢性移植肾肾病(CAN)的关系.方法:用免疫组织化学技术和计算机真彩色图像分析系统半定量检测30例CAN患者移植肾组织中ILK、Ⅳ型胶原和TGF-β1的表达情况,分析三者间及与CAN病理分级之间的关系.10例正常肾组织作为对照.结果:CAN移植肾组织中ILK、Ⅳ型胶原、TGF-β1的表达比正常肾组织明显增加(P<0.001),并随CAN病理分级呈逐渐递增的趋势.移植肾组织中ILK的表达与TGF-β1、Ⅳ型胶原呈正相关(r分别为0,822、0,715,P<0,001).结论:研究结果显示ILK可能介导TGF-β1促进CAN细胞外基质(ECM)异常沉积发病机制,Ⅳ型胶原的异常沉积是CAN患者移植肾纤维化的重要表现,且ILK在CAN移植肾纤维化进程中起重要作用.

목적:탐토이식신조직중정합소련접격매(ILK)급Ⅳ형효원적표체,급여전화생장인자-β1(TGF-β1)표체화만성이식신신병(CAN)적관계.방법:용면역조직화학기술화계산궤진채색도상분석계통반정량검측30례CAN환자이식신조직중ILK、Ⅳ형효원화TGF-β1적표체정황,분석삼자간급여CAN병리분급지간적관계.10례정상신조직작위대조.결과:CAN이식신조직중ILK、Ⅳ형효원、TGF-β1적표체비정상신조직명현증가(P<0.001),병수CAN병리분급정축점체증적추세.이식신조직중ILK적표체여TGF-β1、Ⅳ형효원정정상관(r분별위0,822、0,715,P<0,001).결론:연구결과현시ILK가능개도TGF-β1촉진CAN세포외기질(ECM)이상침적발병궤제,Ⅳ형효원적이상침적시CAN환자이식신섬유화적중요표현,차ILK재CAN이식신섬유화진정중기중요작용.
Objective:To investigate the expressions of integrin- linked kinase(ILK) and collagen Ⅳ in the renal allografts of patients with chronic aUograft nephropathy (CAN), and explore their relationship with transforming growth factor-β1(TGF-β1) ex-pression and the pathogenesis of CAN. Methods: Immunohistochemical assay and computer- assisted genuine colored image analysis system were used to detect the expressions of ILK,TGF-β1and collagen Ⅳ in the renal aUografts of patients with CAN. The asso-ciation between TGF- β1 ,collagen Ⅳ and ILK,as well as the relationship between their expressions and the pathological grading of CAN,were analyzed. Ten specimens of healthy renal tissue were used as controls. Results: The expressions levels of ILK, TGF - β1and collagen Ⅳ were significantly higher in the renal tissues of the patients, compared to those in the normal renal tissues(P<0.001) ,and the expressions tended to increase with the pathological grades of CAN. In CAN group, tbe expression of ILK was posi-tively correlated with those of TGF - β1 and collagen IV ( r = 0. 822 and r = 0.715, respectively; P < 0.001 for both ). Conclusion: It is indicated by the data that ILK might mediate the mechanism through which TGF- β1 promoted the abnormal deposition of ECMin renal aUograft with CAN and the abnormal deposition of collagen Ⅳ in renal aUngraft might be important manifestation of fibrosis in CAN. It is suggested that ILK might play an important role in the progression of renal fibrosis in CAN.
