目的 探讨胃黏膜下层胰岛移植的可行性.方法 应用雄性Wistar-Furth大鼠,STZ介导糖尿病大鼠模型.同种同基因大鼠体外胰岛分离纯化,分别移植进入糖尿病大鼠胃黏膜下层或肾被膜下,移植后不同时间段评估移植物生存率与代谢功能.结果 移植后早期移植胰岛被有效确认,并迅速分泌胰岛素逆转大鼠高血糖状况,糖耐受功能良好,与肾被膜下移植组比较,差异无显著意义.移植后4周,胃黏膜下移植组胰岛移植物胰岛素分泌功能逐渐丧失.结论 胃黏膜下胰岛移植技术具有良好的开发前景,如何提高其远期效果有待进一步考证.

목적 탐토위점막하층이도이식적가행성.방법 응용웅성Wistar-Furth대서,STZ개도당뇨병대서모형.동충동기인대서체외이도분리순화,분별이식진입당뇨병대서위점막하층혹신피막하,이식후불동시간단평고이식물생존솔여대사공능.결과 이식후조기이식이도피유효학인,병신속분비이도소역전대서고혈당상황,당내수공능량호,여신피막하이식조비교,차이무현저의의.이식후4주,위점막하이식조이도이식물이도소분비공능축점상실.결론 위점막하이도이식기술구유량호적개발전경,여하제고기원기효과유대진일보고증.
Objective To investigate the possibility that islets transplanted into the gastric submucosal layer of diabetic rat. Methods Male Wistar-Furth rats were used in the experiment. Diabetes was induced by the administration of streptozocin (STZ). Islets were isolated and purified in syngeneic rats. Islets mass was transplanted into the submucosal layer of the stomach. The graft survival and metabolic function were e-valuated in different phases. Results All STZ induced diabetic rats receiving 1500 islets in the submucosal layer of stomach returned to normoglycemia in 14 d after the transplantation. Histological examination of grafts showed numerous well-granulated insulin-containing cells in the submucosal layer. Compared with the renal subcapsular space transplantation group, no differences were found in the glucose tolerance changes. At the end of follow-up, hyperglycemia was observed in the rats of gastric submucosal transplant group. Conclu-sion The submucosal compartment of the upper GI tract is a viable site, which offers a safe and convenient place for transplantation islet in rodents.
