目的:通过显微形态观察和功能检测分析超抗原(SEA)对Jurkat T淋巴细胞的影响.方法:应用CCK-8法,原子力显微镜(AFM)体外检测Jurkat T淋巴细胞增殖活性、及其细胞膜超微结构的改变.结果:与对照组比较,不同浓度的SEA随着作用时间的增加,细胞膜超微结构与Ra值明显发生改变.Jurkat T淋巴细胞经过质量浓度为0.1-100 mg/L SEA,作用48 h期间,Jurkat T细胞的表面超微结构(平均粗糙度,Ra)和增殖活性(A值)变化非常明显.其中,质量浓度10 mg/L SEA作用24 h,Jurkat T淋巴细胞表面结构变化最明显,而细胞增殖活性在48 h达最强.结论:SEA作为超抗原作用于Jurkat T淋巴细胞后,Jurkat T淋巴细胞的活化表现出表面形态结构改变先于代谢水平的改变,较后者更灵敏.

Aim:To study the effect of superantigens (SEA) on the T cell (Jurkat cell)by observing morphology and functions. Methods: Cell counting kit-8 (CCK-8), atomic force microscope(AFM) was used to check up the proliferation and membrane ultrastructure of T cell. Results: Compared to the un-treated cell, the cellular membrane ultrastructure and Ra value was significantly changed. The T cell treated with SEA(0.1~100 mg/L) for 48 hours, membrane ultrastructure Ra and proliferative activity was changed obviously. The T cell treated with 10 mg/L SEA for 24 hour, the changes of membrane ultra-structure were more remarkable, and at 48 hour the proliferation of T cell was more active. Conclusion: SEA as a superantigens, which treated T cell, and the activation of the T cell showed the morphology changes earlier than the metabolic changes.
