目的研究卵巢肿瘤组织中谷胱甘肽S转移酶-π(GST-π)的表达及其临床意义。方法用免疫组化链霉菌素亲生物素过氧化酶法(S-P法)测定10例良性卵巢肿瘤、40例恶性肿瘤组织中GST-π的表达,及其与组织学类型、病理分级、临床分期及术前化疗的关系。结果10例良性肿瘤GST-π无阳性表达,40例恶性肿瘤阳性表达26例(65%);GST-π的表达与组织学类型和术前化疗无关系(P>0.05)。GST-π阳性表达在低分化癌中占85.71%,高于高、中分化癌(33.33%,P<0.01);在上皮性肿瘤Ⅰ~Ⅱ期中占20%,Ⅲ~Ⅳ期占71.4%,有随着分期升高而升高的趋势。GST-π阳性者缓解期短,易近期内复发。结论免疫组化测定GST-π在卵巢恶性肿瘤中的表达对判定化疗疗效,克服耐药、估计预后具有一定的临床价值。GST-π阳性者属非P-糖蛋白耐药,化疗疗效及预后差。
目的研究卵巢腫瘤組織中穀胱甘肽S轉移酶-π(GST-π)的錶達及其臨床意義。方法用免疫組化鏈黴菌素親生物素過氧化酶法(S-P法)測定10例良性卵巢腫瘤、40例噁性腫瘤組織中GST-π的錶達,及其與組織學類型、病理分級、臨床分期及術前化療的關繫。結果10例良性腫瘤GST-π無暘性錶達,40例噁性腫瘤暘性錶達26例(65%);GST-π的錶達與組織學類型和術前化療無關繫(P>0.05)。GST-π暘性錶達在低分化癌中佔85.71%,高于高、中分化癌(33.33%,P<0.01);在上皮性腫瘤Ⅰ~Ⅱ期中佔20%,Ⅲ~Ⅳ期佔71.4%,有隨著分期升高而升高的趨勢。GST-π暘性者緩解期短,易近期內複髮。結論免疫組化測定GST-π在卵巢噁性腫瘤中的錶達對判定化療療效,剋服耐藥、估計預後具有一定的臨床價值。GST-π暘性者屬非P-糖蛋白耐藥,化療療效及預後差。
목적연구란소종류조직중곡광감태S전이매-π(GST-π)적표체급기림상의의。방법용면역조화련매균소친생물소과양화매법(S-P법)측정10례량성란소종류、40례악성종류조직중GST-π적표체,급기여조직학류형、병리분급、림상분기급술전화료적관계。결과10례량성종류GST-π무양성표체,40례악성종류양성표체26례(65%);GST-π적표체여조직학류형화술전화료무관계(P>0.05)。GST-π양성표체재저분화암중점85.71%,고우고、중분화암(33.33%,P<0.01);재상피성종류Ⅰ~Ⅱ기중점20%,Ⅲ~Ⅳ기점71.4%,유수착분기승고이승고적추세。GST-π양성자완해기단,역근기내복발。결론면역조화측정GST-π재란소악성종류중적표체대판정화료료효,극복내약、고계예후구유일정적림상개치。GST-π양성자속비P-당단백내약,화료료효급예후차。
ObjectiveTo study the expression of glutathione S-transferase-π(GST-π) gene in the tissues of ovarian tumors and its clinical significance. MethodsGST-π gene expression in tissues of 10 benign tumors and 40 malignant tumors was detected by immunohistochemical method of labeled streptavidin biotin(SP method).Chi-square test was used to analyze the interrelation between the expression of GST-π gene and histological type,pathological grade, clinical stage and preoperative chemotherapy. Results Positive expression of GST-π gene was detected in 26 cases out of 40 malignant ovarian tumors(65%); no positive expression was demonstrated in 10 benign ovarian tumors. There was no interrelation between expression of GST-π gene and histological types of tumor tissues and preoperative chemotherapy (P>0.05). The positive expression rate in lowly differentiated cancer (85.71%)was higher than that in highly and moderately differentiated cancer (33.33%,P<0.01), whereas positive expression rate of epithelial ovarian carcinoma in stage Ⅰ~Ⅱ and stage Ⅲ~Ⅳ were 20% and 71.4%,respectively. The expression rate might increase with the progress of tumor stage. The remission period of positive expression group was shorter than that of negative group, and the tumors were easily recurrent in recent days. ConclusionsDetection of GST-π gene expression rate shows certain clinical significance in the judgement of chemotherapy efficacy, overcoming of drug resistance and estimation of prognosis. Patients with positive GST-π gene expression are poor in chemotherapy efficacy and prognosis,for this positive expression belongs to drug resistance mediated by non-P-glycoprotein mechanism.
