利用活性炭的选择吸附性,以活性炭为吸附剂提取玉米浸泡水中的脂多糖.研究了活性炭用量以及浸泡水的pH值对脂多糖提取率的影响,结果表明:活性炭用量为原料重的2%,溶液pH值为6.5最有利于脂多糖的提取;脂多糖从玉米浸泡水中提取出来后,采用活性炭色谱法和离子交换色谱法分离提纯.活性炭色谱法提纯脂多糖的回收率为82.34%,初步产品中还含有较多的蛋白质和普通多糖,在冷冻干燥后再通过离子交换色谱法进一步分离提纯,然后冷冻干燥并脱盐,最终脂多糖产品纯度达到了97.4%.

Lipopolysaccharide was extracted from soak water of maize by active carbon with ability of selective absorption. We studied the effects of dosage of active carbon and the pH value of the soak water of maize on the extraction rates of lipopolysaccharide. The results showed that the best conditions for lipopolysaccharide extraction from soak water of maize are dosage of active carbon should be 2% of the raw material by weight and the pH value of the soak water should be 6.5.After extracted from the soak water of maize, lipopolysaccharide was purified by active carbon chromatogram-separating and ion-exchange chromatogram-separating. The recovery rate of lipopolysaccharide of active carbon chromatogram-separating was 82.34%. After the active carbon chromatogram-separating, we got primary product and it contains some proteins and amylases, need to be purified more by ion-exchange chromatogram-separating. Finally the products were freeze-dried and the salts in it were got rid of. The final product contains 97.4 percent of lipopolysaccharide.
