Ginkgo biloba%flavonoids%orthogonal design
本试验比较了有机溶剂浸提法和超声波法提取总黄酮的效果,选择提取率较高的超声波法来提取。对料液比、超声时间和乙醇体积浓度对银杏中总黄酮提取效果的影响进行了单因子试验,并采用正交试验法找出最佳提取条件为料液比lg:30mL、超声时间30min、乙醇浓度70%。采用紫外可见分光法检测黄酮类化合物的含量,不加显色剂,直接以258nm作为最大吸收波长。结果表明,银杏中总黄酮的平均含量稳定在2.77%,精密度的RSD为0.302%,该方法的平均回收率为100.3%,本试验方法样品处理简单,准确度高,精密度好,适合于银杏叶中总黄酮的提取和测定。

This study discussed organic solvent extraction and ultrasonic extraction method. The extraction ratio of ultrasonic method was higher than the organic solvent extraction method when it was used for extraction of flavonoids. Single factor experiments were done with different extraction conditions including the ratio of Ginkgo biloba and ethanol, ultrasonic thne and the volume fractions of ethanol. The optimum extraction conditions of flavonoids from Ginkgo biloba were investigated by using orthogonal design.The results showed that lg:30 mL,70% ethanol and extraction for 30 min was the optimal condition. Quantitative analysis of flavonoids was achieved by ultraviolet spectrophotometry and do not need add colour - display reagent, detemfined at 258 nm directly as maximum absorption wavelength. The results indicated that the average content of flavonoids in Ginkgo biloba was 2.77 % under the optimal conditions, the relative standard deviation in precision test was t3. 302% ,and the average recovery rate was 100.3%. This study suggested that this method was simple, accurate, and also suitable for the extraction and quantitative analysis of tlavonoids in Ginkgo biloba.
