目的:应用PT-PCR技术和免疫组织化学染色显示人胚胎下丘脑褪黑素受体及其亚型的分布.方法:取人胚胎下丘脑组织,抽提总RNA并应用RT-PCR方法检测其mRNA, 扩增所得阳性产物用自动测序仪测序;石蜡包埋下丘脑组织,应用鼠抗人mt1和MT2受体单克隆抗体分别检测褪黑素mt1和MT2受体亚型mRNA及其蛋白的表达.结果和结论:人胚胎下丘脑组织中存在mt1和MT2受体亚型mRNA的表达及其蛋白的翻译,提示在该组织中有mt1和MT2两种受体亚型的表达.
目的:應用PT-PCR技術和免疫組織化學染色顯示人胚胎下丘腦褪黑素受體及其亞型的分佈.方法:取人胚胎下丘腦組織,抽提總RNA併應用RT-PCR方法檢測其mRNA, 擴增所得暘性產物用自動測序儀測序;石蠟包埋下丘腦組織,應用鼠抗人mt1和MT2受體單剋隆抗體分彆檢測褪黑素mt1和MT2受體亞型mRNA及其蛋白的錶達.結果和結論:人胚胎下丘腦組織中存在mt1和MT2受體亞型mRNA的錶達及其蛋白的翻譯,提示在該組織中有mt1和MT2兩種受體亞型的錶達.
목적:응용PT-PCR기술화면역조직화학염색현시인배태하구뇌퇴흑소수체급기아형적분포.방법:취인배태하구뇌조직,추제총RNA병응용RT-PCR방법검측기mRNA, 확증소득양성산물용자동측서의측서;석사포매하구뇌조직,응용서항인mt1화MT2수체단극륭항체분별검측퇴흑소mt1화MT2수체아형mRNA급기단백적표체.결과화결론:인배태하구뇌조직중존재mt1화MT2수체아형mRNA적표체급기단백적번역,제시재해조직중유mt1화MT2량충수체아형적표체.
Objective:To identify the melatonin receptor subtype(mt1 and MT2) gene and protein distribution in human hypothalamus.Methods:RT-PCR technique and immunohistochemistry method were applied to study the distribution of melatonin receptor subtype gene and protein in the hypothalamus of human embryos,respectively.Results:The electrophoresis of RT-PCR products with mt1 and MT2 subtype revealed specific expression of both melatonin receptor subtypes in human embryonic hypothalamus.Nucleotide sequence analysis indicated that the RT-PCR products coincided with cDNA sequence from human GenBank.The mt1 and MT2 receptor subtypes with immunohistochemistry method was principally localized to cytoplasm and membrane in the hypothalamus.Conclusion:It is possible that mt1 and MT2 subtype of melatonin receptors co-exist in the hypothalamus of the human embryos.
