目的 体外实验初步探讨叶下珠醇提物对人胃癌细胞MKN28生长的影响.方法 通过集落形成试验观察叶下珠醇提物对人胃癌细胞MKN28集落形成的影响;将不同浓度叶下珠醇提物作用于MKN28细胞,倒置显微镜下观察细胞形态变化,MTT法测定叶下珠醇提物对MKN28细胞的增殖抑制率;流式细胞仪检测叶下珠醇提物作用48 h后MKN28细胞的凋亡情况.结果 在2mg/mL和1 mg/mL2个浓度下叶下珠醇提物对MKN28胃癌细胞集落形成具有不同程度的抑制作用,浓度大者.抑制作用更强.MTT法测定细胞增殖抑制率结果表明,叶下珠醇提物对MKN28细胞增殖有抑制作用,其抑制效应具有浓度和时间依赖性.流式细胞检测提示叶下珠醇提物能诱导人胃癌细胞MKN28发生凋亡.结论 叶下珠醇提物能抑制人胃癌细胞MKN28增殖,凋亡是其作用机制之一.

OBJECTIVE To carry out in vitro experiment to investigate into the effect of phyllanthus urinaria alcohol extract on the growth of human stomach cancer cell MKN28. METHOD Colony formation test was adopted to observe the effect of MKN28, MTT method was used to determine its multiplication- inhibiting rate, and flow cytometry was used to detect MKN28 cell apoptosis 48 hours after phyllanthus urinaria alcohol extract took, effect. RESULT Under two different concentrations (2 mg/mL and 1 mg/mL), the extract had different degrees of inhibiting effect on MKN28 colony formation, and the higher the concentration, the better the inhibiting effect. MTT method determination showed that the extract had inhibiting effect on the multiplication of MKN28 cells and the inhibiting effect was dependent on concentration and time. The result of flow cytometry suggested that the extract could induce MKN28 apoptosis. CONCLUSION The alcohol extract of phyllanthus ruinaria can inhibit the growth of human stomach cell MKN28, and apoptosis is one of its mechanisms.
