目的 观察不同浓度的同型半胱氨酸对人脐静脉内皮细胞(HUVEC)作用不同时间后对细胞增殖和形态结构的影响.方法 在含有不同浓度的同型半胱氨酸培养环境及对照培养环境中,分别孵育内皮细胞24 h、48 h、72 h后,采用四甲基偶氮唑盐(MTT)比色法检测同型半胱氨酸对内皮细胞增殖活性的影响,并进行细胞形态学观察.结果 同型半胱氨酸低浓度时对细胞生长的抑制不明显,随着浓度的增高,细胞生长抑制越明显,且呈浓度、时间依赖性.结论 高同型半胱氨酸可损伤内皮细胞,抑制细胞生长,有助于糖尿病和胰岛素抵抗致动脉粥样硬化等血管病变的发生和发展.

목적 관찰불동농도적동형반광안산대인제정맥내피세포(HUVEC)작용불동시간후대세포증식화형태결구적영향.방법 재함유불동농도적동형반광안산배양배경급대조배양배경중,분별부육내피세포24 h、48 h、72 h후,채용사갑기우담서염(MTT)비색법검측동형반광안산대내피세포증식활성적영향,병진행세포형태학관찰.결과 동형반광안산저농도시대세포생장적억제불명현,수착농도적증고,세포생장억제월명현,차정농도、시간의뢰성.결론 고동형반광안산가손상내피세포,억제세포생장,유조우당뇨병화이도소저항치동맥죽양경화등혈관병변적발생화발전.
Objective To investigate proliferation and morphological structure of human umbilical vein endothelial cells (HUVEC) incubated with homocysteine.Methods Endothelial cells were incubated in 1640 cultivate medium with different concentrations of homocysteine and the control cultivate medium for 24 h,48 h,and 72 h.Then cell proliferation activity and cell morphology were detected by MTT colorimetric assay.Results The cell growth inhibition was not obvious in low concentrations of homocysteine.As the concentrations were increased,cell growth inhibition was more obvious dose-dependently and time-dependently.Conclusion Hyperhomocysteinemia could damage the endothelial cells,inhibit cell growth,and contribute to the occurrence and development of atherogenic vascular disease of diabetes and insulin resistance.
