本研究通过富勒烯与赖氨酸直接反应,合成了一种富勒烯赖氨酸衍生物(C_(60)-Lys),并通过元素分析、红外光谱、~1H-NMR、~(13)C-NMR等对其分子结构进行了表征.细胞毒性研究发现C_(60)-Lys对人淋巴母细胞AHH-1的毒性较小,在1200 mg/L浓度以下的细胞存活率和未用药对照组相比无明显差异.辐射生物效应研究发现400 mg/L以上C_(60)-Lys预处理能显著提高受照细胞的存活率,并降低辐射诱发的凋亡率,其辐射保护效应具有明显的剂量依赖性.说明C_(60)-Lys预处理对受4Gyγ射线照射的AHH-1细胞具有一定的保护作用.

In this study,we synthesized a fullerene derivative C_(60)-Lys via direct reaction of fullerene(C_(60))and Lysine.whose molecular structure was identified by elemental analysis,infrared spectrum,1H-NMR and ~(13)C-NMR.It has been found that the chemical C_(60)-Lys has little toxicity on human lymphoblast cell AHH-1.CCK-8 assay and Annexin V/PI staining assay Were performed to evaluate the cell survival rate and apoptosis after γrays irradiation.The cell survival rate of C_(60)-Lys treatment groups(≤1200 mg/L)shows no significant difference compared with the control group.The results show that pretreatment of cell with C_(60)-Lys(≥400 mg/L)markedly increases the cell survival and decreases the cell apoptosis rate after irradiation exposure in a dose-ependant manner.These results can be suggested that C_(60)-Lys has certain radioprotection effects to AHH-1 cell against radiation induced damages.
